B-lymphocyte stimulator (BlyS) is a member of the tumor necrosis factor family that is known to take part in B cell activation, proliferation, and differentiation, and play an important role in the non-specific immunity and specific immunity. However, studies on BlyS in teleosts are very limited, especially in golden pompano (Trachinotus ovatus). In this study, we conducted in vivo as well as in vitro functional analyses of a BlyS homologue from the golden pompano. (i) Analyze the expression of BlyS under normal and pathogen infection conditions; (ii) Mutate the key amino acids of BlyS and clarify the immune activation mediated by BlyS; (iii) Reveal the relationship between BlyS and TLRs in immune defense against pathogens through overexpression and RNAi; (iv) Explore the potency as vaccine adjuvant for fish. The project will provide insights on the fuction and mechanism of BlyS against pathogens infection in golden pompano, and may exploit vaccine adjuvant for the control of pathogens infection in fisheries.
B淋巴细胞刺激因子(BlyS),属于肿瘤坏死因子(TNF)家族,与B淋巴细胞存活、增殖、分化及抗体分泌关系密切,在机体非特异性及特异性免疫中发挥着重要作用。鱼类BlyS基因的抗病机制研究较哺乳动物相对匮乏。为此,本项目拟以我国华南地区重要深水网箱养殖鱼类卵形鲳鲹为研究对象,在前期相关研究基础上,采用分子生物学、免疫学和细胞学等手段,研究BlyS基因结构和表达特征;分析BlyS的表达和调控规律;探讨BlyS生物学活性及免疫作用机制;综合利用干扰表达、过量表达及转染等技术,在细胞(卵形鲳鲹吻细胞系)和个体水平上系统分析卵形鲳鲹BlyS介导的非特异性及特异性免疫机制,解析BlyS参与的信号通路,揭示BlyS参与卵形鲳鲹抗病原感染的主要调控机制;并进一步探索卵形鲳鲹BlyS作为佐剂在鱼类病害免疫防治中的应用前景。本项目将有助于了解卵形鲳鲹的免疫机制,为鱼类疾病免疫防治提供新的理论依据和思路。
B淋巴细胞刺激因子(BLyS)是肿瘤坏死家族中的重要成员,能促进B细胞生长、分化和凋亡,且对T细胞有一定的调节功效,在哺乳动物的免疫系统中起着重要作用,但是,在硬骨鱼中对BLyS功能的研究相对匮乏。本研究以卵形鲳鲹(Trachinotus ovatus)为对象,克隆并鉴定了卵形鲳鲹BLyS基因 (TroBLyS),并且分析了其序列结构特点,对TroBLyS基因体内和体外的功能进行了初步研究。qRT-PCR结果表明,TroBLyS在健康卵形鲳鲹的11个组织中均有表达,且在头肾中表达量最高,其次是脾脏和肝脏;在哈维氏弧菌刺激后,TroBLyS在肝脏、脾脏、头肾中表达量显著增加。体外重组蛋白TroBLyS(rTroBLyS)可以促进头肾淋巴细胞的增殖和抑制淋巴细胞的凋亡。体内实验表明,TroBLyS在卵形鲳鲹体内过量表达显著促进了巨噬细胞的活化,增加了细胞因子和趋化因子的分泌,并减少了哈维氏弧菌在组织中的感染量;当卵形鲳鲹体内TroBLyS的表达被其siRNA干扰时,卵形鲳鲹肝脏、脾脏和肾脏组织中细菌的感染量显著增加。确定BLyS123位点,186-189位点对其活性的影响。此外,在哈维氏弧菌感染下,注射了2周pTroBLyS质粒的鱼成活率显著高于对照组,且pTroBLyS质粒组免疫基因的表达量也显著提高。该研究表明,TroBLyS在卵形鲳鲹的抗细菌免疫中发挥了重要作用,并有具有作为免疫增强剂的潜能。
