Rise in the incidence of thyroid tumor and younger trend, the early diagnosis of thyroid cancer have become a difficult clinical problem due to the mechanisms is unclear. The effect of screening new cancer related gene molecular markers, the development has the tumor targeting of macromolecular contrast agent, is of great significance to early diagnosis of thyroid tumor. Our present study have first time found that SHP-2 high expression in the tissue of the solid tumors of thyroid carcinoma, and to promote the malignant biological behavior of thyroid cancer cells; SHP-2 over-expression of Nthyori-3-1 cells more likely to undergo malignant transformation and that the nude mice of SHP-2 over-expression obviously increase the tumorigenicity.SHP-2 may to promote thyroid cancer progression. This study intends to by has established SHP-2 transgenic nude mice model and cell model, Synthetic dual-modality with ultrasound and magnetic targeted contrast agent, to establish an in vivo imaging evaluation system for accurate detection of early thyroid cancer.
甲状腺肿瘤目前发病率呈上升和年轻化趋势,由于发病机制不明,使甲状腺癌早期诊断成为临床亟待解决的难题。筛选新的肿瘤发生相关基因的分子标记物,开发具有肿瘤靶向性的高分子造影剂,对甲状腺肿瘤早期诊断意义重大。我们前期研究首次发现SHP-2高表达于实体肿瘤甲状腺癌组织中,并促进了甲状腺癌细胞的恶性生物学行为;同时发现SHP-2高表达的Nthy-ori 3-1细胞更易发生恶性转化,且SHP-2高表达的裸鼠,其致瘤性明显增加;提示SHP-2可能参与促进甲状腺肿瘤发生。本研究拟合成超声/磁共振双模式靶向造影剂,用前期构建的SHP-2裸鼠模型及细胞模型,拟建立活体评估甲状腺癌的影像评价体系,实现其早期准确诊断。
甲状腺癌目前发病率呈明显上升趋势,缺乏有效的早期检测手段,确诊主要依靠细针穿刺病理学检查,但是对于较小的早期肿瘤细针穿刺敏感性不高,容易漏诊,这迫切要求探索一种新的甲状腺癌诊断方法。分子影像学可对活体在分子或细胞水平上进行定量分析研究的新技术,能够克服传统成像手段评估肿瘤时存在缺乏特异性、难以定量等缺点,为肿瘤的研究引入了新的思维方式和研究手段。我们前期研究证实,SHP2是一种甲状腺癌特异性表达的酪氨酸磷酸酶,在甲状腺癌组织中特异性高表达,是甲状腺癌的敏感标志物。因此,用相应的 SHP2抗体制备分子探针有可能实现甲状腺癌的特异性成像,达到早期诊断的目的。.乳酸/羟基乙酸共聚物(Polylactic-co-glycolic acid,PLGA)是由乳酸和羟基乙酸随机聚合而成,是一种可降解的高分子化合物,具有良好的生物相容性、无毒、可在体内完全降解为二氧化碳和水,已经通过 FDA 认证。PLGA 上的基团可进行进一步的修饰,医学领域应用最为广泛的是 PLGA-COOH。分子探针上可结合顺磁性或超顺磁性物质来缩短纵向弛豫时间或横向弛豫时间,从而在 MR 仪下进行显像。因此,本研究拟利用 PLGA-COOH 纳米粒为载体,内部包裹液态氟碳, 在纳米粒表面连接 SHP2 抗体及顺磁性对比剂 Gd3+,构建SHP2靶向甲状腺癌的特异性MR/超声双模态分子探针,并探讨该分子探针读甲状腺癌细胞的靶向特异性及其超声/MR成像的可行性在体内和体外实验。
