Morinda officinalis How. mainly distributes in South China, its dried fleshy root is one of the famous traditional Chinese medicine with great medicinal value, such as invigorating kidney, strengthening immunity and anti-depression. Many researches focused on pharmacological functions and chemical compositions have been done, however, there is a severe shortage of studies about gene of Morinda officinalis How., especially about genome and transcriptome. Gene markers to quickly assess quality or to distinguish counterfeits have not been mined. To expand understanding and promote industrialization, total mRNAs from Morinda officinalis How. samples will be sequenced using the Next Generation Sequencing platform. Based on de novo assembly and comparative transcriptomics, genes profile in stem, leaf, root and seed will be showed. Genes show quite concordant with changes of nystose will be identified, from which Morinda officinalis How. specific gene markers to fast evaluate the content of nystose will be discovered. A database about Morinda officinalis How. will be constructed. This project may select gene markers to fast estimate quality and distinguish counterfeits, may find SSR markers to discriminate origins. 巴戟天以根入药,为我国四大南药之一,是重要的传统中药材,具有补肾壮阳、增强免疫力和抗抑郁等多种功效。目前对巴戟天的研究主要集中于药理功能、化学成分等方面。基因水平的研究却比较少,尤其缺乏基因组和转录组信息,也没有快速鉴定巴戟天品质优劣及辨别混伪品的分子标记。为了更好的理解巴戟天的生长发育机理与促进巴戟天的产业化发展,本项目拟利用二代测序技术对巴戟天多组织多样本进行高通量测序,基于转录组从头拼接与比较转录组学的方法,系统解析巴戟天茎、叶、根、种子等组织的基因表达谱;鉴定与巴戟天药用成分(耐斯糖)协同差异的基因,挖掘可快速检测药用成份含量的巴戟天特异的分子标记;建立巴戟天专题数据库。本项目的顺利实施,有望挖掘出能快速区分品质良莠及混伪品的分子标记,鉴定出辨别产地的遗传标记,从而为巴戟天的选育工作与产品分级提供指导,为利用基因工程技术改良巴戟天提供数据基础。
