Cyrtotrachelus bugueti is one of the major boring-insects, which makes the control measures difficult relatively and caused devastating loss, to many kinds of caespitose bamboos. Besides, C. bugueti is not only a harmful insect, but also a resource insect. And the chemistry control of C. bugueti individually could not control the insect or exploit it at the same time, so the analysis of information-oriented chemical of mating and molecular perception mechanism of C. bugueti and make use of the host's sex attractant to tentakill, or collect, the insect is one of the most important way to control the insect. The study will choose C. bugueti, which always caused serious harm in the southern part of China, as the experimental material. In this study we will measure the EAG and behavioral responses of C. bugueti adult to the volatiles of polypide, and then analyse the synergism, which are made by the orientation of the host material to the volatiles of polypide, afterwards we will clone and express the pheromone-binding protein of adults, finally we will anlayse the combinative status between pheromone-binding protein and active components. Besides, we will analyse the enticing results of sex attractant in forest to declare the perception mechanism of sex pheromone of adults, and then we will obtain the chemical fingerprinting of the adult distinguish the volatiles, and declare the the synergism, which are made by the orientation of the host material to the volatiles of polypide, and finally we will explore the the technology and method, which could control the population of C. bugueti by use the sex attractant.
竹笋象甲是多种丛生竹笋的重要钻蛀性害虫,由于防治困难,常造成毁灭性损害。而且竹笋象甲既是有害昆虫也是资源昆虫,单纯的化学治理不能兼具防治与利用两个方面的利益,研究成虫觅偶定位信息化学物质及其分子感受机制,利用性引诱剂实施诱杀(集)是防治该类害虫的重要途径。本项目以我国南方发生普遍、危害严重的长足大竹象为研究对象,通过测定成虫对虫体挥发物成分的EAG和行为反应;研究寄主植物VOCs对虫体挥发物的增效作用;克隆、表达成虫PBPs;分析PBPs对活性组分的结合情况;测定利用性引诱剂在林间对成虫的引诱效果,从而明确长足大竹象成虫性信息素感受机制;破解成虫识别虫体挥发物的化学指纹图谱;揭示寄主定向信息物质对虫体挥发物的增效作用;探索利用性引诱剂控制长足大竹象种群的方法和技术。
长足大竹象Cyrtotrachelus buqueti是我国南方重要的钻蛀性林业害虫,危害多种丛生竹类,严重影响竹类产业发展。在昆虫世代繁衍过程中,雌雄个体间如何通过嗅觉系统远距离精确搜索定位成为雌雄个体间是否有效交流的关键。本研究观察了长足大竹象交配、产卵行为,使用吸附管吸附法动态收集雌虫性信息素,采用GC-MS鉴定了性信息素成分与日变化节律,利用Y-型嗅觉仪与触角电位仪分析了对成虫的行为影响。在分子生物学方面,利用RT-PCR技术成功克隆得到长足大竹象的一个性信息素结合蛋白基因(Genbank登录号:KU845733.1),利用SYBR Green 1荧光染料法,采用荧光定量PCR分析了CbuqPBP1在雌雄成虫不同组织中的转录表达谱,并进行表达纯化,运用荧光竞争结合技术对该蛋白与主要虫体挥发物和寄主挥发物等结合特性进行分析,研究了该蛋白的功能。
