To investigate the change of the expression level of obese gene and the serum concentration of Leptin in NIDDM diabetics rats,and to research the effect of different intensities of exercise on the expression level of obese gene and the serum concentration of Leptin in NIDDM diabetics rats,to demonstrate how exercise decrease the weight and improve the disorder of glucose metabolism. Methods 20 OLETF rat and 15 LETO rat are randomly allocated to four groups: non-exercise OLETF group, exercise OLETF group, non-exercise LETO group and exercise LETO group.After 9 weeks of exercise on 20 OLETF rats and 15 LETO rats, the level of ob gene expression is detected by fluorogenic quantitative RT-PCR method. the plasma concentration of Leptin and insulin are measured by means of radioactive-immune kit. Results The level of obmRNA expression and the plasma level of Leptin in non-exercise OLETF groups are significantly higher than non-exercise LETO groups(P<0.01), The level of obmRNA expression and the plasma level of Leptin in exercise OLETF groups are significantly lower than non-exercise OLETF groups after nine weeks of exercise(P<0.01). there is not a direct relationship between the fall of Leptin's plasma concentration and exercise enhancing insulin sensitivity in OLETF rats.Conclusion these changes about obese gene's expression level and Leptin's concentration of OLETF rats may be a self-regulation which can maintain the balance of energy metabolism.
本课题观察运动对Ⅱ型糖尿病大鼠脂肪细胞肥胖基因表达和血清瘦素水平的影响,以及对血糖、血清胰岛素等生化指标的影响,研究运动减轻Ⅱ型糖尿病大鼠体重,改善糖代谢紊乱的可能机理;通过观察不同运动量对糖尿病大鼠上述指标及糖代谢的影响,探讨糖尿病运动的适宜运动量,为临床上开展糖尿病的运动疗法提供指导。
