Nucleoprotein UHRF1 is highly expressed in a variety of tumors, such as renal cell carcinoma (RCC). It has become a hot topic in tumor epigenetics, but the mechanism of UHRF1 in RCC needs to be studied deeply. The results of the preliminary study showed that down-regulation of UHRF1 expression in RCC could significantly inhibit cell proliferation and promote cell apoptosis, and it was found that TXNIP was significantly up-regulated in UHRF1 down-regulated cells and simultaneous reduction of TXNIP can save the phenotype caused by the reduction of UHRF1 alone. Accordingly, UHRF1 is likely to play a role by direct regulation of TXNIP. This project intends to further study the molecular mechanism of UHRF1 in regulation of TXNIP. The HIF-1 binding site was found in the UHRF1 promoter region, and it was found that of HIF-1 and UHRF1 expressions showed consistency in clinical specimens. We intend to further expand the sample size to verify our hypotheses, and use feasible epigenetics research methods to study the relationship between HIF-1 and UHRF1, so as to reveal the mechanism and signal pathway of UHRF1 in RCC, and discover the new molecular target of renal cell carcinoma, which has important scientific significance and research value.
核蛋白UHRF1在肾癌等多种肿瘤中特异性高表达,已经成为肿瘤表观遗传学的一个研究热点,但UHRF1在肾癌中的作用机制有待深入研究。本项目组前期研究发现:下调肾癌细胞UHRF1表达,可显著抑制细胞增殖并促进其凋亡,利用基因芯片技术并筛选验证发现:下调UHRF1细胞中TXNIP显著上调,如同时下调TXNIP可挽救UHRF1下调所引发的表型变化,从而推断UHRF1很可能通过对TXNIP直接调控而发挥作用。本项目拟在此基础上进一步深入研究,明确UHRF1调控TXNIP的分子机制;UHRF1启动子区域存在HIF-1结合位点,且预实验发现临床标本中HIF-1与UHRF1表达呈一致性,我们拟进一步扩大样本量进行验证,并应用可行的表观遗传学研究方法,研究HIF-1与UHRF1的相互关系,从而较全面的揭示UHRF1在肾癌中的作用机制和信号通路,以期发现新的肾癌分子靶点,具有重要的科学意义和研究价值。
核蛋白UHRF1在肾细胞癌等多种肿瘤中特异性高表达,已经成为肿瘤表观遗传学的一个研究热点,但其在肾细胞癌中的作用机制还不清楚。我们的前期研究发现:通过siRNA技术下调UHRF1表达可在体外显著抑制肾癌细胞的增殖和侵袭,并引起细胞凋亡;利用基因芯片技术并筛选验证发现:下调UHRF1细胞中TXNIP显著上调,且同时下调UHRF1和TXNIP可挽救UHRF1单独下调所引起的细胞生物学表型,在此基础上,我们对UHRF1调控TXNIP表达的具体机制进行了深入研究。我们利用ChIP实验检测发现UHRF1可直接结合于TXNIP基因的启动子区,而下调UHRF1的表达可下调UHRF1的结合,并同时下调该区域组蛋白去乙酰化酶HDAC1的结合水平,并且该区域H3K9的乙酰化水平显著升高,提示UHRF1可能通过对TXNIP启动子区H3K9的乙酰化修饰而调控其表达,从而调控肾癌细胞的增殖、凋亡和侵袭。通过本项目研究,我们明确了UHRF1在肾癌中发挥了癌基因功能,首次明确了乙酰化修饰分子UHRF1在肾癌中的表达情况及其对肾癌细胞增殖、凋亡和侵袭的重要调控作用和潜在的分子调控机制,为将UHRF1作为肾癌诊断的分子标志物以及针对UHRF1的肾癌靶向治疗的研究和靶向药物开发提供了理论依据和体外实验基础。
