Function Dyspeps(FD) is a common and frequently-occurring disease,which affects the living quality of patients seriously.The medication treatment does not achieve an ideal effect and it is hot clinic issue needed to be solved immediately.Recently,a large number of evdence-based medicine proved that FD belongs to level Ⅰin acupuncture indications'spectrum and it has an exact clinic effect.However,the related mechansim has not been expounded clearly yet.The lattest reserch showed that:the crucial link for FD occurance lies in the dysfunction of ICC-GSMC functional net induced by braingut petide content , its receptor expression and the change of SCF/c-Kit signal transduction pathway.Hence,this project ,under the guidence of meridians , collaterals and acupoints of acupuncture theory,taking the FD rats model stimulated by clipping the tails as the object,observe the regulation function of electroacpuncture on gastrointestinal motility,gastrointestinal functional net with different frequencies,at the point of view of braingut petide Ghrelin,which affect the crucial link of gastrointestinal motility ,ICC-GSMC functional net ,and the closely-related SCF/c-Kit signal transduction pathway,applying histomorphopogy,immunohistochemistry,in situ hybridization conbined with computer image analysis,real-time fluorescent quantitative RT-PCR and other techniques,to discover the pathway and mechanism of acupuncture treating this disease,in this way,to provide a powerful evidence for the popularization of advantaged diseases of acupuncture.
功能性消化不良(FD)是消化系统的常见病、多发病,严重影响患者生活质量,药物治疗不理想,是目前亟待解决的临床热点问题。近年来大量循证医学证据证实FD属于Ⅰ级针灸病谱的范畴,针灸治疗临床效应确切,然而相关的作用机制尚未阐释清楚。最新的研究表明:脑肠肽含量变化及其受体表达异常与SCF/c-Kit信号转导通路受损所引起的ICC-GSMC功能网络失调可能是FD发病的关键环节。故本项目以针灸经络腧穴理论为指导,以夹尾刺激的FD大鼠模型为研究对象,从影响FD胃肠动力关键环节的具有代表性的脑肠肽Ghrelin、CCK和ICC-GSMC功能网络、以及密切相关的SCF/c-Kit信号转导通路入手,采用组织形态学、免疫组化、原位杂交结合计算机图像分析、实时荧光定量RT-PCR等技术,观察电针对模型大鼠胃肠动力、胃肠功能网络的调整作用,拟揭示针灸治疗该病的作用途径和机制,为针灸优势病种的推广应用提供坚实的依据
