Developing authentic genuine material medica is a reliable way to solve the problem of drug quality control and lacking of resources. The base is the scientific recognition of the unique quality characteristics of authentic genuine material medica, as well as the deep investigation of the mechanisms of their quality formation. As a typical often used material medica with multiple derivatives, the quality of Gentiana Radix is very complicated. At the same time, as a staple material medica with remarkable special genuine characteristics and many production areas, Gentiana Radix is in huge demand. This causes the urgent question of its resources. Gentiana Radix from Shaanxi as a well known authentic genuine material medica has an important quality characeristic of high content of secoiridoids glycosides, which is the principle active constituent of this crude drug. However, its quality formation mechanism is still not clear. This may be the main obstacle of raising the quality and supply of cultivated Gentiana Radix. In this study, Gentiana macrophlla from Shaanx is adopted as the object, using two research systems of cultivation and in vitro tissue culture to systemetically investigate the rules of the accumlation of main active constituents. The rules of exprression of three key genes covering the whole scope of the principle active constituents biosynthesis in Gentiana macrophlla. will also be deeply studied on three aspects of time, space and induced by stresses. Furthermore, the inner formation mechanisms of the major quality character of authentic genuine Gentiana Radix from Shaanxi, that the high content of iridoid mainly represented by gentiopicroside will be preliminaryly explored on the basic of synthetical correlation analysis of those two kinds of rules mentioned above.The results of quality formation mechanism study of Gentiana Radix from Shaanxi will contribute to studies of formation mechanisms of authentic genuine material medica, the most challenge and attractive area in material medica studies, providing a typical case and idea. It can be used in GAP of Gentiana Radix in Shaanxi, instructing the establishment of SOP, keeping and improving the quality of cultivated Gentiana Radix. So, this study has both important theoretical and practical values.
发展道地药材是解决中药材质量和资源问题的可靠途径,其基础是对道地药材质量的科学认识和对其质量形成机制的深入了解。秦艽是典型的多来源和有明显道地性的大宗常用药材,质量问题复杂,需求量大而迫切,资源问题突出。陕西秦艽是优质道地药材,具有以龙胆苦苷为代表的裂环环烯醚萜含量高的质量特征。目前,对其质量形成机制了解甚少是严重阻碍秦艽药材质量提高和资源供应的瓶颈。针对这种现状,本课题以陕西秦艽为对象,采用栽培和离体两种研究体系,对其主要有效成分积累变化规律进行系统研究,并且对有效成分合成途径中上、中、下游最关键的三个关键酶基因表达规律从时间、空间和环境因素诱导作用三个方面进行系统考察。在两类规律相关性综合分析的基础上,探讨道地陕西秦艽质量形成的内在机制。研究结果可为道地药材形成机制研究提供思路和例证,具有理论意义;成果应用于指导秦艽规范化生产,稳定和提高药材质量,提供大量优质药材,具有积极实践意义。
