Phosphodiesterase4(PDE4) plays an important role in regulating the levels of cAMP in airway smooth muscle and inflammatory cells. PDE4 is not only used as the molecular target for anti-asthma drugs, but also a hotspot in the development of new anti-asthma drugs abroad. The aim of our project is to make preparations for the large scale of filtering PDE4 inhibitors by constructing a yeast strain of high performance and persistent ,stable expressing of human PDE4A . With the use of the DNA reconstruction technology, a large amount of PDE4 can be expressed and used in the research of PDE4. Under the support of the project fund, a series of studies on PDE4 and PDE4 inhibitors were also carried out successfully.
磷酸二酯酶4(PDE4)是调节气道平滑肌和炎症细胞中cAMP水平的重要因素。本项目通过DNA重组技术建立高效持续稳定表达人PDE4A的酵母细胞株。此酵母细胞株的建立可用于大量产鶳DE4A,经过纯化的PDE4A可用于PDE4研究。本项目为建立PDE4B、C和D酵母细胞株和为建立大规模筛选PDE4抑制剂的方法奠定基础。.].
