The Study on Mechanism of Xiao-Long -Tong-Bi Capsule Reducing the Volume of Prostate Jia Jinming, Jiao Yongzheng Guang Anmen Hospital, Academy of Chinese Traditional Medicine, Beijing 100053 Abstract: Objects: On the clinical background of reducing the volume of prostate of benign prostatic hyperplasia (BPH) with the traditional Chinese medicine Xiao-Long -Tong-Bi Capsule (XLTB ), the mechanism of decreasing the size of prostate in BPH is explored in this research. Methods: The male rats castrated and were injected of testosterone propionate were applied as animal models. With the technology of flow cytometry, in situ end-labeling(ISEL),immunohistochemistry quantitative analysis and RT-PCR, the effect of XLTB Capsule on the epithelial cell division cycle,basic fibroblast growth factor(bFGF),nitricoxide synthase(NOS) ,bcl-2 protein and its mRNA expression ,the epithelial cell apoptosis in prostatic gland. Results: Compared with those in the model group,the prostate volume and the odds of glands weight/body weight in the high dose group with XLTB were obviously low (P<0.01),the G2+M period of epithelial cells in this group was obviously longer.The express level of bFGF in rats prostate fed with high dose of XLTB is much lower than that in the simple model rats(P<0.01),and the expressions were predominantly located in the epithelial cytoplasm,seldom in nuclei and stroma.The NOS was mainly histochemically localized to nerve fibers coursing through the smooth musculature of the stroma. After 3 weeks,the length density of NOS-containing nerves in the prostate with XLTB was much higher than that in other three groups(P<0.001).XLTB and finasteride caused a peak of of apoptosis on day 7 ,a less on day 14 and a little recover on day 21 in epitheial cell of prostate in normal rats. The distribution patten of bcl-2 in rats' prostateis is the same as that of bFGF. The bcl-2 protein and its mRNA expression in the two doses groups of XLTB are both fewer than those in BPH model group(P<0.01). Conclusions: XLTB could inhibit the expressing of bFGF in prostatic tissue,subsequently inhibit the mitoses of the epithelial cells in prostate of BPH rats.In addition,this agent could increase the NOS-containing nerves and restrain the expression of bcl-2 and its mRNA in the prostate of BPH rats, all which result in accelerating the apoptosis of epithelial cells in rats prostate prostatic tissus.All these probabely may be the mechanism of shrink the prostate volume in BPH treating with XLTB..

本项研究采用免疫组化定量技术,基因转录及流式细胞双波长分光光度及观测手段,探索消癃通闭通过对细胞增殖、分裂及BfGF、NO的作用治疗前列腺增生的机理。该研究将对中医中药治疗前列腺增生开拓一条新路,与国际相关研究接轨,并进一步揭示前列腺增生的发病机理。具有重大的社会意义、学术意义及经济效益。.
