Eukaryotic CDKs are a group of cyclin-dependent protein kinases, which function in diverse cytological processes, including regulations in cell cycle progression, transcription, splicing and transcription. Their kinase activities are controlled in several ways. Our previous results showed that Arabidopsis CDKG1 interact with a splicing factor RSZ33, to regulate the pre-mRNA splicing of a pollen wall related gene, CalS5, thus participate in the regulation of pollen wall synthesis. However, the CDK activity regulation for CDKG1 is not yet known, and only one downstream gene was found controlled by CDKG1. We hope to find the protein factors regulating CDKG1 activity and other possible CDKG1 might be involved, using molecular biology methods. And we hope to find more downstream genes and the function model of CDKG1, via RNA sequencing for mutant cdkg1 and RNA immuno-precipitation assay for CDKG1-GFP cdkg1 trans-gene lines. This research will expand the knowledge about theoretical basis of CDK function mode at molecular level, and has the application prospect in agricultural production, as CDKG1 regulates pollen development.
真核生物的CDK是一类周期蛋白依赖的蛋白激酶,它们种类多样,功能各有不同,有些调控细胞周期的进程,也有些CDK在转录、剪接和翻译等细胞学过程中发挥作用,它们的活性受到多种方式的调控。我们此前的研究表明,拟南芥CDK家族的一个成员CDKG1能和剪接因子RSZ33 相互作用从而调控一个花粉壁合成相关基因CalS5前体mRNA的剪接,进而参与到花粉壁合成的调控过程。然而该CDK的活性调控方式尚不清楚,受其控制的下游基因也只发现一个。本项目拟采用分子生物学研究方法,找到和CDKG1相互作用的其它蛋白因子并进而发现CDKG1是否参与剪接之外的其它生物学过程以及CDKG1蛋白激酶活性的调控方式;获取更多的CDKG1的下游基因并分析CDKG1作用模式。本项目的研究可以在分子水平上拓展CDK作用模式的理论基础,由于CDKG1与花粉发育的关系,又具备在农业生产上应用的前景。
