The interactions and subsequent effects between various ions and human serum albumins (HSA) or bovine serum albumins (BSA) have been systematically studied by means of equilibrium dialysis, ultraviolet-visible (UV-Vis), fluorescence, circular dichroism (CD) and Raman spectroscopy. These ions include essential elements such as I-, Mn2+, Fe2+, Co2+, Ni2+; rare earth element La3+; toxic element Cd2+; the potential medicinal ions such as Pd2+ Ag+, Au3+, phosphotungstic acid. Furthermore, the competitive binding of bimetal ions, including Cu2+ and Fe3+, Cu2+ and Zn2+, Cu2+ and Mn2+, Zn2+ and Mn2+, Zn2+ and Hg2+ to serum albumins also has been intensively investigated in the multicomponent complex systems. The stepwise stability constants of metal ions binding to serum albumins are obtained with non-linear least squares method fitting Bjerrum formula. The affinities of various metal ions for the binding sites in serum albumins have been evaluated, and the synergstic effect and anatagonism occurring in the binding processes have also been discussed. In addition, the dynamics rate constants and thermodynamics parameters are also been calculated to characterize the interaction mechanisms and subsequent effects. The detailed studies are contributed significantly for understanding the structure (conformation) and the physiological behaviors of serum albumins, which provide the necessary theory for developing new medicines to maintain the balance of trace metal ions.
通过平衡透析、分光光度法和电化学等方法研究两种金属离子与血清白蛋白形成的三元体系,用非线性最小二乘法拟合得出金属离子与血清白蛋白结合的逐级稳定常数值;探讨不同种类金属离子对血清白蛋白的亲和力以及不同金属离子与血清白蛋白相互作用的协同或拮抗行为;用动力学方法结合热力学平衡研究结果探讨金属离子与血清白蛋白结合的机理及后续效应。通过本项目的研究可加深对血清白蛋白的结构(构象)和生理功能的认识,为寻找调整人和动物体内金属离子水平的新药提供指导。
