Cataracts are the first blindness disease worldwide and responsible for nearly 50% of the total blind. The research indicated that cataract is relative with oxidative damage. Heme oxygenase-1 is an important anti-oxidative defensive enzyme, to date the relation between HO-1 and cataract is unknown. In our previous study we have got FVB transgenic mice expressing HO-1 mutant HO-1G143H through microinjection technique. During the breeding we found that the transgenic mice got cataract at the age of about four mouths, meanwhile the negative control mice was normal. The findings indicate that HO-1 may play an important role in the generation and development of cataract. This study will first elucidate the mechanism of the development of cataract in the transgenic FVB mice expressing HO-1 mutant HO-1G143H , then we will try to reveal the correlation of HO-1 and cataract in the lens epithelium cells. Our study will provide not only the fully information of the role of HO-1 in the generation and development of cataract, but the theoretical and experimental foundation in the therapy of the cataract using HO-1.
白内障是世界范围内的首位致盲疾病,在世界盲人总数中近百分之五十是因白内障所致。研究表明白内障的形成与氧化损伤有极大的相关性。血红素加氧酶-1(heme oxygenase-1,HO-1)是机体中存在的一种重要的抗氧化防御酶,到目前为止,HO-1与白内障发生、发展的相关性还不甚明确。在前期研究中我们通过显微注射技术建立了表达HO-1突变体HO-1G143H的FVB转基因小鼠,经过饲养繁殖我们发现该转基因小鼠在16周龄左右会患有白内障,同批生长的阴性小鼠则正常,这个现象提示我们HO-1在白内障的发生、发展中发挥重要作用。因此,本研究拟首先对HO-1突变体HO-1G143H FVB转基因小鼠白内障发病机制进行分析,进而在晶状体上皮细胞系中对HO-1与白内障相关性进行阐明。本研究不仅为解析HO-1在白内障发生发展中的作用提供更全面的资料,也为临床白内障治疗中的HO-1策略提供理论基础与实验依据。
