Studying the compatibility characteristics and clinical application rules of TCM has great theoretical significance and practical value for improving the level of clinical treatment, developing the theory of compatibility of TCM and creating modern Chinese medicine. Salvia miltiorrhiza - safflower is the classic representative medicine for activating blood and removing stasis, and is widely used in cardiovascular and cerebrovascular diseases. However what is the optimal combination of the effective group of Salvia miltiorrhiza - safflower group against cerebral ischemia reperfusion (I/R) damage, there has no research report yet. Some pathological links of the cerebral I/R injury to the project (free radical injury, inflammatory reaction and apoptosis), combined with the neurological score, infarct volume, serum SOD, LDH, MDA, IL-1 β,IL-6 content, Bcl-2 and Caspase-3 mRNA in the hippocampus of ischemic side, using the corresponding surface method (RSM) with elitist non dominated sorting genetic algorithm (NSGA-Ⅱ) and entropy TOPSIS method for quantitative visualization, safflower group (Salvia Danshensu, salvianolic acid B, protocatechuic aldehyde and hydroxysafflor yellow A) combined withthe multi-objective optimization. Taking drug pair as the breakthrough point, this article is of great significance for revealing the scientific connotation of prescription compatibility, mining and improving the compatibility theory of Chinese medicine compound, and developing the thought of clinical medication.
研究药对的配伍特点和临床应用规律,对提高临床治疗水平、发展中医药配伍理论和创制现代中药具有十分重要的理论意义与实践价值。丹参-红花是经典代表性活血化瘀相须药对,广泛应用于心脑血管疾病。但丹参-红花有效组分群抗脑缺血再灌注(I/R)损伤物质基础的最优组合是什么,尚无研究报道。本项目拟针对脑I/R损伤的某些病理环节(自由基损伤、炎性反应、细胞凋亡等),结合神经功能评分,梗死体积,血清SOD、LDH、MDA、IL-1β、IL-6含量,缺血侧海马区Bcl-2、Caspase-3 mRNA值等指标,采用相应曲面法(RSM)结合带精英策略的非支配排序遗传算法(NSGA-Ⅱ)和熵权TOPSIS法可视、定量的进行丹参-红花组分群(丹参素、丹酚酸B、原儿茶醛和羟基红花黄色素A)配伍的多目标优化。本项以药对为切入点,对于揭示方剂配伍的科学内涵,挖掘和提高中医复方配伍理论,开拓临床用药思路具有十分重要的意义。
