基于AGEs蓄积研究中医外治“掺药法”促进糖尿病大鼠溃疡愈合机制

Diabetic foot ulcer is difficult to cure, the wound tissue AGEs accumulation and related growth factor function disorder is an important factor affecting wound healing. Clinical research through the traditional Chinese medicine external treatment of diabetic foot ulcers, Chinese medicine can effectively promote the granulation tissue proliferation, accelerating wound healing. This paper focuses on " diabetic foot ulcer - the accumulation of AGEs - doped concentration method - to improve the cell biological activity to promote wound healing"the scientific hypothesis represented by traditional Chinese medicine " medicine mixing method " using the external treatment of drug " Yinhuang powder " as the breakthrough point, through the treatment of diabetic foot ulcers, using immunohistochemistry, real-time fluorescent quantitative PCR method and Western blot method, changes of AGEs and TNF-a, IL-1, bFGF, EGF, VEGF, PDGF and other factors of wound tissues, promote the relevant mechanism of diabetic foot ulcer healing and action links from the tissue, cellular and molecular level of external therapy of traditional Chinese medicine " medicine mixing method ", to provide to help the healing process and the influence factors to further clarify the diabetic foot ulcer wound.

糖尿病足溃疡属难愈性溃疡的范畴，其创面组织AGEs蓄积及相关生长因子功能紊乱是影响创面愈合 的重要因素。通过中药外治糖尿病足溃疡的临床研究发现，中药可以有效促进创面肉芽组织增生，加速创面的愈合。本课题围绕"糖尿病足溃疡- AGEs蓄积-掺药法-提高细胞生物学活性-促进创面愈 合"科学假说，拟以中医外治"掺药法"的代表外治药物"蚓黄散"为切入点，通过治疗糖尿病足 溃疡，应用免疫组织化学、实时荧光定量PCR法及Western blot等方法，观察创面组织中AGEs及TNF -a、IL-1、bFGF、EGF、VEGF、PDGF等因子的变化，从组织、细胞和分子层面探讨中医外治"掺药法"促进糖尿病足溃疡愈合的相关机制与作用环节，为进一步明确糖尿病足溃疡创面的愈合过程和影响因素提供帮助。
