Hepatocellular carcinoma (HCC) is one of the most malignancy and the leading cause of human cancers. Although encouraging long-term survivals of HCC patients have been obtained in some clinical centers, HCC carries a very poor prognosis and the 5-year survival rate is limited to 25-39% after surgery. One of the major reasons of the poor prognosis is its high rate of recurrence and metastasis after surgery. The other is that early detection of HCC remained very unsatisfactroy. Therefore, it is urgent to identify more HCC-associated molecules which will be helpful for developing novel target genes for diagnosis and therapy. In this project, we isolated a novel HCC-associated gene MXR7. We found that this protein was undetectable in normal adult liver and other tumors, however it was highly expressed in HCC especially in AFP negative HCC (73.17%), suggesting it is a potential candidate for HCC diagnosis. Using gene reconstruction technology, we produced the antigen produced in protokaryotic or eukaryotic cells and prepare several specific antibodies. Especially, we successfully established six cell lines that can highly manufacture monoclonal antibodies against MXR7. Further than that, we investigated its application in HCC by serology through developing a kit for HCC ealy diagnostic assay that is further supported by "863" program. In China, we applid for one patent for MXR7 monoclonal antibody which will be another helpful diagnostic tool for HCC in coadjutant with AFP.
米托恩醌耐药相关基因MXR7是新近发现的在肝癌内特异性最好的高表达基因。本研究拟建立血清以及基因诊断技术,检测肝癌患者血清中该蛋白的含量以及外周血中微量播散的肿瘤细胞,探讨该方法的敏感性与特异性,作为肝癌的早期诊断指标和早期血液转移诊断指标。该研究对肝癌患者的预后判断以及选择适当的治疗方案均具有极其重要的意义及价值。
