Angiogenesis plays a vital role during the process of pressure ulcer healing, which provides nascent tissue with oxygen and nutrients constantly. Recent study found micro electric field can make vascular endothelial cell proliferation, migration,improving its survival and permeability,thus promote angiogenesis of pressure ulcers. However,the mechanism of the above study is still unclear. Base on the theory of BangCi acupuncture in Traditional Chinese Medicine, and combine with clinical effects of eletro acupuncture on bedsores,this study puts forward a new idea of BangCi electroacpuncture enhancing angiogenesis of pressure ulcers .The Molecular biology, Western blot,Immunohistochemistry, RT-PCR and TUNEL will be used to detect the area of pressure ulcer, local blood flow,micro vessel density, the expression of VEGF, VEGF-2 and the effect of the protein and mRNA expression of sign signal molecules in the signaling pathways. This will help clarifying the effect of promoting angiogenesis of bedsores with BangCi electroacupuncture;elucidating the mechanism of the possible signal pathway of vascular endothelial cell proliferation, migration,improving its survival and permeability of angiogenesis in bedsores with BangCi electroacupuncture;revealing the mechanism of enhancing angiogenesis of bedsores with BangCi electroacupuncture;exploring the new ways to treat pressure ulcers, which provide experimental and theoretical evidence for treating pressure ulcer, and chronic wound healing.
血管新生在褥疮组织修复过程中起着至关重要的作用,它不断给新生的组织提供氧气和营养成分。近期研究发现微电场可以使血管内皮细胞增殖、迁移、提高其存活和通透性,从而促进褥疮组织血管新生,但其作用机制还不完全清楚。本研究在中医傍刺针法理论指导下,结合针刺治疗褥疮和微电场促进褥疮组织修复的显著疗效,提出电针傍刺法促进褥疮组织血管新生的新理念。我们拟采用分子生物学、免疫组化、Western blot、RT-PCR和TUNEL等技术检测褥疮组织面积、局部血流、微血管密度和VEGF、VEGFR-2 及其信号通路中标志性信号分子的蛋白和mRNA 表达;明确电针傍刺促进褥疮血管新生的作用;阐明电针傍刺促进褥疮组织血管新生过程中参与血管内皮细胞增殖、迁移、存活及渗透性的可能信号通路;揭示电针傍刺法促进褥疮组织血管新生的机制,探索治疗褥疮的新途径,为临床电针傍刺修复褥疮,乃至慢性损伤组织愈合提供理论和实验依据。
