Peptide nucleic acid (PNA) was one of the most prominent analogues of nucleic acid. In order to understand the special recognition between PNA and DNA, much space information about the complex of PNA and DNA should be obtained. In this project, three new peptide nucleic acids (PNAs) and their complementary DNAs were synthesized and purified. The hybridization of the PNAs with DNAs were investigated by using UV spectroscopy and NMR spectroscopy. The NMR spectroscopy of an octomer PNA were partially assigned preliminarily, and the formation of base pairs between the PNA and DNA were observed in NMR spectra.
我们设计合成了一类全新结构的肽核酸(PNA),具有序列特异性识别靶DNA并形成螺旋结构的能力。我们拟采用核磁共振(NMR)法解析此螺旋的三维结构,并结合其它研究方法鱌NA与DNA分子识别的结构基础。由于DNA在生物体内的重要生理功能,本研究结果将对从核酸水平人为调控功能基因的表达以及发展作用于核酸的药物都具有指导意义。因此具有学术价值和对应用的潜在指导意义。
