Effectors could induce or suppress plant defense responses in plant–microbe interactions. The identification and functional characterization of the plant pathogen effector is one of the most important themes in plant pathology study, and will promote our understanding of the pathogenic mechanism of phytoplasmal disease. The Chinese jujube (Zizyphus jujube Mill.) is a widely grown and popular fruit tree in China. Jujube witches’-broom (JWB) disease is a destructive disease caused by Jujube witches’-broom phytoplasma and seriously hinders the development of the jujube industry. Because of the inability to culture phytoplasmas in cell-free media and the lack of screening tools of phytopalsma effector, the research progress of the function and mechanism of effector in the pathogenicity of phytoplasmas become more slowly. In this project, we will analyze the mixed transcriptome of phytoplasma infecting Chinese jujube using the RNA-Seq technique, and screen the phytoplasma coding sequence (CDS) for candidate effector. Then we will determine the type of the phytoplasma effector using transient and stable transformation, and analyze their function in plant development and growth. The expected results will further our understanding on the molecular mechanism of phytoplasma pathogenicity and explore novel ways for the phytoplasmal disease control.
植物病原菌效应因子能抑制植物的防卫反应,是植物与病原菌互作研究中的热点问题。效应因子的鉴定和功能分析能促进人们对植物病原菌致病机制的了解。枣疯病是枣树上危害最严重的植原体病害,目前没有有效的防治措施。植原体不能体外培养,致使效应因子的筛选受到限制,植原体致病机理的研究也陷入瓶颈。本项目拟利用高通量RNA-Seq技术对感病植株进行转录组学分析,预测植原体效应因子的编码蛋白框,利用瞬时表达和稳定转化分析植原体效应因子的类型,并对其影响植物发育的代谢通道进行预测,探讨效应因子在枣疯植原体致病过程中的功能及作用机制。本项目在理论上可以分析效应因子在枣疯植原体致病性方面的功能和作用机制,深化对枣疯病致病机理的了解,在实践上可为探索有针对性的防控策略提供科学指导。
植物病原菌效应因子能抑制植物的防卫反应,是植物与病原菌互作研究中的热点问题。效应因子的鉴定和功能分析能促进人们对植物病原菌致病机制的了解。枣疯病是枣树上危害最严重的植原体病害,目前没有有效的防治措施。植原体不能体外培养,致使效应因子的筛选受到限制,植原体致病机理的研究也陷入瓶颈。本项目通过高通量测序获得了枣疯植原体基因组的完成图,筛选到枣疯植原体候选效应因子57个。对枣疯植原体的imp基因进行了原核表达并制备了抗血清。免疫胶体金标记表明金颗粒特异性的标记到了感病枣树叶片中脉、茎、根中植原体细胞的外膜上。本项目在理论上可以分析效应因子在枣疯植原体致病性方面的功能和作用机制,深化对枣疯病致病机理的了解,在实践上可为探索有针对性的防控策略提供科学指导。
