Lycium barbarum polysaccharide is the major component derived from Traditional Chinese Medicine of Lycium barbarum L. It is well documented that lycium barbarum polysaccharide has antioxidant property,and exerted protective effects on lipid peroxidation,and hepatic,myocardial and retinal injury etc.Our previous study demonstrated that lycium barbarum polysaccharide has retinoprotective effects aganist retinitis pigmentosa animal model via preservation of nuclear transcriptional factor NF-κB.Therefore, in the present study,to obtain more experimental data for further clinical application of lycium barbarum polysaccharide on retinitis pigmentosa as a new drug, the protective effects of lycium barbarum polysaccharide on rat retinal degeneration are to be investigated by the combined methods of pharmacology, molecular biology,histopathology and biochemistry.The inhibitory effects of lycium barbarum polysaccharide on N-methy-N-nitrosourea-induced retinal apoptosis will be observed,and its molecular mechanism on modulation of NF-κB pathway are to be investigated, including regulation of cytoplasmic and nuclear NF-κB level;status of poly (ADP-ribose) polymerase (PAPR), caspase and oxidative stress which activation of NF-κB .
枸杞多糖是宁夏特色药食两用道地中药材枸杞的主要成分,具有抗氧化作用,对脂质过氧化、肝损伤、心肌损伤和视网膜损伤等均具有保护作用。我们前期研究结果显示:枸杞多糖对视网膜色素变性损伤动物模型具有保护作用,其机制与调控视网膜细胞核转录因子NF-κB有关。为此,本课题以N-甲基-N亚硝脲致大鼠视网膜变性损伤为模型,综合采用药理学、分子生物学、病理学和生物化学等方法与技术手段,进一步研究枸杞多糖对大鼠视网膜变性损伤的保护作用及其分子机制,重点检测其对视网膜细胞凋亡的抑制作用;对NF-κB信号通路的调控作用:包括对NF-κB在细胞浆和核水平的影响;对激活NF-κB的多聚腺苷二磷酸核糖聚合酶(PARP)、Caspase的活化和氧化应激反应状态。为枸杞多糖成为临床防治视网膜色素变性损伤的新药提供实验依据。
目的:本研究探讨了枸杞多糖对N-甲基-N亚硝脲(MNU)致大鼠视网膜变性损伤的保护作用及其机制。方法:雄性Sprague-Dawley大鼠,按60mg/kg/只,腹腔注射MNU建立视网膜变性损伤动物模型。枸杞多糖(100, 200和400mg/kg)于大鼠注射MNU前7天始,灌胃给药。分别于大鼠注射MNU 24小时和7天后,采集视网膜和血清标本,进行视网膜变性损伤病理观察,视网膜细胞调亡检测,NF-κB/p65,IκBα 和caspase3, 7, 9等蛋白表达western blot测定,以及血清氧化应激生物标志酶、血脂、肝和肾损伤标志酶等的检测。结果:枸杞多糖可抑制MNU致大鼠视网膜的变性损伤和细胞凋亡。Western blot检测结果显示,视网膜细胞核NF-κB p65含量,在大鼠注射MNU 24小时后升高,7天后降低,而胞浆IκBα 含量和Caspase3, 7, 9的表达,在大鼠注射MNU 24小时和7天后,均升高;枸杞多糖可显著升高视网膜变性损伤大鼠视网膜细胞核NF-κB p65的含量,并使胞浆IκBα含量以及caspase3, 7, 9的表达恢复至正常水平。枸杞多糖还可增加大鼠注射MNU 7天后血清总抗氧化能力(T-AOC),超氧化物歧化酶 (SOD) 和谷胱甘肽过氧化物酶(GSH-PX) 的活性,并抑制脂质过氧化物(MDA)和一氧化氮(NO)的含量。此外,枸杞多糖对MNU所致大鼠血脂、肝和肾损伤标志酶等的改变有一定影响。结论:枸杞多糖对MNU致大鼠视网膜变性损伤具有抑制作用,其机制与枸杞多糖维持视网膜细胞NF-κB水平,抑制caspase3, 7, 9,并提高机体抗氧化能力有关。
