The dry leaves of the Aconitum kusnezoffii Reichb are commonly used as Mongolian medicine, which has many effects, such as relieving pain, clearing away heat and toxic, and inflammation. This scientific subject mainly focus on studying the leaves of the Aconitum kusnezoffii Reichb. Its chemical composition was firstly analysized via spectroscopy, chromatography, and mass spectra methods. Based on the composition analysis, monocytes / macrophages (RAW264.7 cells) were used as inflammatory models in vitro to investigate the effective ingredients of Aconitum leaves. Lipopolysaccharide (LPS) was used as inducer to pre-screen the relavent chemical composition. In addition, flow cytometry was applied to verify the active ingredients, which were responsible for anti-inflammation.The selected active ingredients were further evaluated in vitro via assays according to animal pharmacodynamics and toxicology.On the other hand, the anti-inflammatory mechanism of the selected active ingredients of Aconitum leaves was elucidated by using different molecular pharmacological techniques, including RT-PCR, Western blot, and ELISA,the mechanism of anti-inflammation and chemical structures of selected active ingredients of Aconitum leaves will be confirmed by this research project,and this project will supply the scientific basis for futher study on Aconitum leaves.
草乌叶为毛茛科植物北乌头Aconitum kusnezoffii Reichb.的干燥叶。具有清热解毒、止痛、杀粘等功效,为常用蒙药。本课题拟以草乌叶为研究对象,通过相应的化学方法提取,采用现代光谱、色谱、质谱等分析手段,分析检测其化学成分,运用体外培养单核/巨噬细胞系(RAW264.7细胞)并由脂多糖(LPS)诱导建立的炎症模型对化学成分进行高通量筛选,初步筛选其具有抗炎作用的药效组分;采用基于流式细胞仪微球阵列的炎性因子高内涵分析技术对其具有抗炎作用的药效组分进行复筛,确认具有抗炎作用的有效成分;结合药效学实验和毒理学实验,对其抗炎有效成分进行评价;运用实时定量RT-PCR、Western Blot、ELISA等技术,对草乌叶具有抗炎作用的有效成分进行深入研究,以期阐其抗炎作用机理并确定其有效化学结构,为草乌叶的深入研究提供科学依据。
