Depression is caused by a variety of factors together, the underlying neurobiological mechanisms are still poorly understood, its prevention and treatment is difficult. We have in deep studied famous TCM doctor's experience and practiced and researched for a few years, and fully proved that Invigorating Qi Kaiyu medicine has definite curative effect by regulation of the neurovascular unit (NVU) and promoting nerve regeneration. On this basis, with combination of epigenetics new progress especially in the study on the regulatory mechanism of non coding RNA, we propose the hypothesis that the mechanism of Invigorating Qi Kaiyu antidepressant is related to the steady state regulation of NVU through lncRNA-miRNA network. By establishing rat model of chronic stress depression and cell models in vitro and the traditional Chinese medicine intervention, using microarray, quantitative PCR, living cells detection technology without labeling, and so on, to observe the lncRNA-miRNA network、NVU and neural regeneration change at the level of whole, cells and genes, to study its non transcriptional gene silencing mechanism for providing a new way of pathogenesis of depression based on the bio psycho social mode; clear the effective key genes and signal pathways of kaixin Jieyu Fang and its effective herbal pair, clarify advantage links or the network of the Invigorating Qi Kaiyu method for controlling NVU micro environment, to reveal scientific connotation of the pathogenesis and treatment of traditional Chinese medicine, to provide new ideas and methods for the prevention and treatment of depression.
抑郁症为多因素共同致病,目前对其发病机制了解有限,防治困难。在深入挖掘名老中医经验,并经实践研究证明益气开郁中药治疗抑郁症疗效确切,能有效调节神经血管单元(NVU)稳态、促进神经再生的基础上,课题组结合表观遗传学特别是非编码RNA调控机制研究新进展,提出益气开郁法疗效机制可能与长链非编码RNA(lncRNA)-微小RNA(miRNA)网络对NVU稳态调控有关的假说。通过建立慢性应激抑郁大鼠模型、体外细胞模型及中药干预,采用基因芯片、定量PCR、无标记活细胞检测等技术,从整体、细胞、基因水平,观察lncRNA-miRNA网络调控、NVU稳态及神经再生改变,研究其非转录基因沉默机制,为生物-心理-社会模式下探索抑郁症发病机制提供新途径;明确开心解郁方及其有效药对的作用关键基因及信号途径,阐明益气开郁法对NVU微环境调控的优势环节,揭示中医病机治法的科学内涵,为抑郁症防治提供新思路和新方法。
对于神经血管单元(NVU)稳态与神经可塑性综合调控研究,特别从中医药中寻求对NVU保护的有效治法和方药成为最有希望的领域之一,而表观遗传学方面的新进展又为生物-心理-社会模式下研究抑郁发病机制提供了新途径,因此长链非编码RNA(lncRNA)、微小RNA(miRNA)网络与NVU及神经再生调控的研究将有望为抑郁防治提供更加有效的新思路和新方法。本课题组在充分继承和深入挖掘名老中医经验基础上,结合前期开心解郁方及其有效药对对改善慢性应激行抑郁模型大鼠的行为学、组织学及其神经再生的研究成果,基于我们认为的抑郁症的核心病机为元气亏虚、气血郁滞和本虚标实证候特征,以及运用开心解郁方治疗该病多年的工作积累,我们认为开心解郁方及其有效药对的作用机制可能与其调控lncRNA、miRNA网络,改善NVU稳态,有效促进神经再生相关。因此我们利用基因芯片技术及生物信息学技术发现了开心解郁方及其有效药对可能调控的lncRNA、miRNA,并通过荧光素酶报告实验验证了其相关性,发现开心解郁方中柴胡的有效成分之一柴胡皂苷d可以调控NF-κB/miR-155/FGF2以发挥其治疗抑郁的作用;同时我们在发现开心解郁方治疗抑郁症时, GNB3在其过程中可能发挥重要的作用,为求进一步的研究其作用方式,我们构建了GNB3基因的高表达和低表达的慢病毒,以备后续深入的研究。同时开心解郁方中及其有效药对可以改善皮质酮损伤原代海马神经元及神经干的形态和凋亡,并可以改善皮质酮损伤后神经干细胞的增殖和分化。本次实验对中药治疗抑郁症的靶点和机制的研究,为中国医药更好的运用于抑郁症的治疗奠定了坚实的理论基础,具有重大的临床意义。
