The abnormal maturation in oocytes will lead to failure in fertilization and embryo development. Mammal oocytes are maintained in meiotic arrest until LH surge. Natriuretic peptide system has been becoming hotspot in oocyte maturation in recent years. We previously reported that BNP plays an important role in porcine oocyte maturation. LH down-regulates BNP and NPR2 expression levels and thus induces oocyte maturation. However, the mechanism of LH in the regulation of BNP and NPR2 expression remains unknown. Moreover, our results indicate that porcine BNP is able to substitute synthetics such as IBMX in the application of synchronizing oocyte maturation. In this study, we will focus on the regulation mechanism of LH on BNP and NPR2 expression and the effect of BNP on oocyte development. The study will help better understanding the mechanism of meiotic arrest in large mammals and provide applied value in production and clinic.
卵母细胞的异常成熟会导致后续的受精及胚胎发育出现障碍。生理条件下,哺乳动物的卵母细胞的发育是先阻滞然后在激素LH诱导下恢复减数分裂而成熟。钠肽系统在哺乳动物卵母细胞减数分裂阻滞中的作用是目前相关研究领域的前沿和热点之一。我们已有的研究表明,BNP在抑制猪卵母细胞成熟中起着重要的作用。LH通过下调BNP及其受体NPR2的表达从而促进猪卵母细胞成熟。然而,LH调控BNP/NPR2表达的具体机制并不清楚。同时,我们的已有的结果提示猪BNP可能作为化学合成物质IBMX等的替代物应用于体外同步卵成熟相关的生产及临床实践。因此,本研究将以体外模型和体内模型相结合,着重开展LH对BNP及其受体NPR2的表达调控研究及这种调控对卵母细胞成熟质量的影响。研究结果有助于人们加深对大型哺乳动物减数分裂阻滞机制的理解。同时,我们的研究在生产和临床上可能具有积极的应用价值。
