Control of tumor metastasis is the key to cancer treatment. P-5m, an octapeptide derived from domain 5 of HKa, was initially found to inhibit the invasion and migration of melanoma cells in vitro and in vivo. P-5m is the shortest sequence reported covered D5 which has anti-metastasis activity similar to D5 full-length sequence. In the previous work, we found that: 1) P-5m inhibited the metastasis of human hepatocarcinoma HCCLM3 cells. 2) P-5m suppressedpulmonary metastasis of hepatocellular carcinoma in vivo. 3) P-5m Inhibited MMP-2 expression in vitro and in orthotopic hepatoma animal model. 4) P-5m inhibited the formation of liver cancer H22 ascites tumor and prolonged the survival time of tumor-bearing mice. On the basis of previous work, this study will investigate the molecular mechanism of inhibition of tumor metastasis by P-5m, define the route of its cell entry, probe the cell signal pathway and molecular network of its inhibitory effect, and find the mechanism of suppression of MMP-2 expression by P-5m for the R&D of P-5m drug.
肿瘤转移是癌症治疗的瓶颈。P-5m八肽来源于高分子激肽原第五结构域(D5),有研究显示其在细胞和整体水平均能抑制小鼠黑色素瘤的转移。这是目前发现的来源于D5的有效抑制肿瘤转移的最短多肽。在前期研究中,我们发现:(1)P-5m显著抑制肝癌HCCLM3细胞的浸润;(2)在人源肝癌裸鼠肺转移模型中,P-5m有效抑制肿瘤肺转移;(3)在整体和细胞水平上,P-5m显著降低肝癌细胞MMP-2的表达;(4)P-5m能够抑制肝癌H22腹水瘤的形成,并延长荷瘤鼠生存期。但P-5m发挥抗肿瘤转移作用的具体分子机制尚不清楚。本项目将在前期工作的基础上,深入探讨P-5m八肽抑制肿瘤转移的分子机制,明确P-5m进入细胞的途径,重点突破P-5m发挥作用的分子网络和有效通路,发现P-5m影响MMP-2表达的分子机制,为后期药物的研发奠定理论基础。
(1)P-5m八肽来源于高分子量激肽原第五结构域。有报道指出,P-5m八肽在细胞水平能够抑制小鼠黑色素瘤细胞的粘附和侵袭,并能抑制小鼠黑色素瘤肺转移,HGK是其关键模序。本课题组近年来围绕P-5m八肽的抗肿瘤转移作用进行研究,前期研究利用细胞水平和整体水平的实验,检测了P-5m八肽对人源肝癌细胞侵袭转移的影响,结果显示P-5m抑制HCCLM3细胞的迁移和侵袭能力,并抑制裸鼠肝癌的肺转移;P-5m对肝癌细胞转移的抑制作用与降低MMP-2表达水平有关。但P-5m发挥抗肿瘤转移作用的具体分子机制尚不清楚。.(2)本项目在前期工作的基础上,通过肿瘤细胞转移相关指标的检测,包括形态学观察和相关基因及蛋白的检测,进一步明确了P-5m对肝癌细胞转移过程中表型的影响;通过利用各种内吞抑制剂等方式对P-5m的穿膜途径进行研究,发现P-5m转运过程与细胞膜网格蛋白和脂筏均有关,P-5m作用后细胞膜表面形成有被小窝;通过分离TH17细胞初步探讨了Th17细胞在P-5m八肽抗肿瘤转移过程中的作用;P-5m与5-Fu联合应用显著抑制H22肝癌模型鼠腹水的生成,显著延长小鼠生存时间,可能与细胞周期阻滞和腹膜渗透性降低有关;为了增加P-5m的稳定性、延长其半衰期、便于临床应用,本研究构建了P-5m–Fc融合蛋白以增强其成药性。本研究为揭示P-5m八肽抗肿瘤转移的机制研究提供了新的实验依据,为抗肝癌转移和癌症腹腔化疗药物的开发提供了新的思路。
