Stylosanthes guianensis, belonging to self-pollination,is an excellent perennial tropical legume forage. Emasculation is difficult as the flower is very small, which seriously hindered the development of the cross breeding. So it is impontant to get stable male sterile line to development the cross breeding. It is significant to search molecular markers which tightly linked to pollen sterility gene on S. guianensis breeding. It not only can be used to judge and discard the undesired materials earlier, but also can be used to screening stable male sterile line, which lay the foundation for S.guianensis cross breeding. In the previous studies. S. guianensis cv. Reyan No.5 was selected as recurrent parent. The BC1F1 plants have been got through continuous cross between S. guianens 1979 (male sterility) and S. guianensis cv. Reyan No.5. In this research, molecular markers linked to pollen sterility character of S. guianensis is going to screening with SRAP and ISSR thchnology in BC1F1 polulation hybrided, then the SRAP and ISSR markers which linked to pollen sterility gene will be conversion to SCAR markers. The purpose is to screening stable male sterile line of S. Guianensis, which lay the foundation for S.guianensis cross breeding.
柱花草属自花授粉植物,花小、去雄非常困难,严重阻碍了其杂交育种的开展,因此获得稳定的雄性不育系是实现这一目标的前提。筛选与柱花草花粉不育基因紧密连锁的分子标记,不仅可以及早判断、淘汰不符合经济性状要求的植株,也可以筛选稳定的雄性不育系,为柱花草杂交育种奠定基础。前期研究中,我们以不育株系1979柱花草为母本,以热研5号柱花草为轮回亲本,获得了BC1F1代可育和不育群体。在此基础上,本项目拟采用SRAP及ISSR技术与BAS法相结合,筛选与柱花草花粉不育基因紧密连锁的SRAP及ISSR分子标记,再将其转化为更稳定而有效的SCAR标记,旨在筛选稳定的雄性不育系,为柱花草杂交育种奠定基础。
柱花草属自花授粉植物,花小、去雄非常困难,严重阻碍了其杂交育种的开展,因此获得稳定的雄性不育系是实现这一目标的前提。筛选与柱花草花粉不育基因紧密连锁的分子标记,不仅可以及早判断、淘汰不符合经济性状要求的植株,也可以筛选稳定的雄性不育系。因此,本研究以雄性不育株系1979柱花草为母本,以热研5号柱花草(花粉可育)为轮回亲本,通过连续套袋杂交,获得了BC1F1代可育和不育分离群体。在此基础上,本项目以BC1F1代分离群体为研究对象,利用SRAP及ISSR分子标记技术结合BAS法筛选与柱花草花粉不育基因紧密连锁的SRAP及ISSR特异分子标记,并将之转化为稳定的SCAR特异标记。本研究筛选获得了与柱花草花粉不育基因连锁的SRAP标记6个、ISSR标记3个;并且成功建立SRAP-SCAR标记6个、ISSR-SCAR标记3个。同时获得了一批优良的可用做育种亲本的柱花草雄性不育种质,为柱花草杂交育种工作奠定良好基础。
