Vagus nerve stimulation (VNS) is one of the effective means of refractory epilepsy treatment, but its action mechanism remains unclear. In many experiments, Galanin has been recognized as an protective neurotransmitter closely related with epilepsy and have impact on the sodium channel. Our previous research has found that after VNS Galanin level in human blood significantly increased, and the degree of increasing has positive correlation with seizure control, but the role of VNS to galanin in the central nervous system and sodium channel, and the role of the change of galanin to sodium channel are not clear.. This study was based on the establishment of rat epilepsy model,then randomized and the establishment of a control group, in which stimulating vagus nerve. Galanin content in brain extracellular fluid of hippocampus is measured by high-performance liquid chromatography, meanwhile, Protein and mRNA content of Galanin and its receptors’ in the hippocampus tissue is measured by Western Blot, real-time PCR method and performed controlled study. In addition, the sodium channel activity in epilepsy rat brain cells after VNS is probed through patch-clamp technique, further testing the role of Galanin's in this process.. Through method described above, on different levels,we attempt to observe the role of VNS on galanin and Na+ channel in hippocampus of epilepsy rat, deeply exploring the mechanism of VNS effect Galanin level, and further control sodium channel, and ultimately achieve the antiepileptic effect..
迷走神经刺激(VNS)是治疗难治性癫痫的有效手段之一,但作用机制不明确。甘丙肽属肽类递质,已证实是与癫痫密切相关的保护性神经递质,并且对Na+通道有调控作用。.我们前期研究发现VNS可使血液甘丙肽水平明显升高,且升高程度与癫痫控制效果正相关,但对中枢神经系统甘丙肽以及Na+通道的作用不明确,甘丙肽的变化对Na+通道的影响也不明确。本研究拟通过建立癫痫大鼠模型,随机分组并设立对照,进行迷走神经刺激,通过液相色谱法测定海马组织细胞外液中甘丙肽含量,同时应用RT-PCR、Western Blot等技术检测海马组织中甘丙肽及其受体的mRNA、蛋白含量,进行对照研究。此外,运用膜片钳技术研究VNS对癫痫大鼠海马细胞Na+通道活性的影响及甘丙肽在其中的作用。.通过从不同层面观察VNS对癫痫大鼠海马甘丙肽以及Na+通道的影响,探索VNS影响甘丙肽进而调控Na+通道,最终达到抗癫痫效果的作用机制。
