A series of non-water soluble and water-soluble porphyrins and metal complexes were synthesized and structure characterized. As mimics of SOD (superoxide dismutase) and CAT(catalase) enzymes, metalloporphyrins were tested in the assays to dismutase O2·-, to decompose H2O2, to clear HO·, and to eliminate lipid peroxidation produced in the homogenated livers of mice. An LKB-2277 thermal activity monitor was used to monitor the cell metabolic activity in vitro to produce themogenesis curves. The growth rate constants k, maximum heat production rates pmax and peak time of growth themogenic curves t of E coli in different metalloporphyrins at different concentrations were deduced from these thermogenesis curves. Some rules were obtained from the biological activity testing results. Among these metalloporphyrins, water-soluble metal porphyrins were most effective.
设计、合成并结构表征几类具有抗活性氧酶功能的新型卟啉及相应不同金属配合物;测定金属卟啉的光谱和电化学氧化还原性质;测试各种金属配合物作为超氧化歧化酶和过氧化氢酶多功能模型物在动物体内外消除多种氧自由基的效力和毒性;探讨模型物的结构与活性关系;初步提出一种体内消除氧自由基化学模型物以指导筛选高效低毒抗多种氧自由基的新药。
