Forensic entomology has unique advantage to resolve the difficulty of determination of death time,which is one of the keynote problem in forensic practice,but whether it can or not give full play to forensic practice is depend on the systemic forensic entomology research in local environment and reliable,convenient molecular species identification technology. .The technology of species identification is based on morphs of insects in past,which restricted the forensic application of entomology because of high-level specialty knowledge and experience needed. In recent years,molecular methods were used in forensic entomology for species identification,but it also cannot fully meet the requirement of forensic practice, so lots of research works were carried out to explore new molecular technology for insect's species identification..It is the first time that systemic researches were carried out for forensic entomology purpose in the unique natural environment of hainan island,and the technology of high resolution melt(HRM)was employed for forensic species identification,which have the advantages of low cost, high-throughput and Specificity,quick and easy operation, it fully meets the special demand of forensic practice. Studies were also launched to explore new generation technology for molecular specie identification and gene bank construction in forensic entomology based on HRM method..Although the important value of HRM in molecular specie identification has been proved in few works,but it is still not been made attention in forensic entomology area until now,so the start of this study should make a history chance to striving for ranking among the world's most advanced in forensic entomology area of china.
死亡时间推断是司法实践需要解决的核心问题之一,法医昆虫学有着独特的优势,其用于实践的关键是应用地法医昆虫学得到系统研究并建立可靠、方便的昆虫种属鉴定技术。.传统的昆虫形态学方法进行种属鉴定对技术、经验要求高,难以在司法实践中推广。近年兴起的分子种属鉴定技术提供了新的思路,但目前在用的技术方法尚难以满足实践需要,探索新的分子鉴定技术是目前研究的关键和热点。.本研究首次对海南岛独特自然环境中法医昆虫学进行系统研究,率先引入全面满足法医学需要的低成本、高通量、高特异性、快速易操作的高分辨溶解曲线技术(HRM)进行种属鉴定,探索建立法医昆虫学新一代分子种属鉴定标准技术方案,构建法医昆虫学分子基因库。.虽然其他领域已初步证明HRM技术进行分子种属鉴定的重要价值,但该技术尚没有被法医昆虫学领域所发现和重视,本研究的开展将为实现法医昆虫学真正运用于法医学实践做出重要贡献。
死亡时间推断作为法医学实践亟待解决的关键问题,法医昆虫学有着独特的优势,特别是在尸体高度腐败的情况下。法医昆虫学应用于司法实践的关键是应用地法医昆虫学得到系统研究并建立可靠、方便的昆虫种属鉴定技术。.本研究以家兔为实验模型,对海南省6个代表区域、四个季度的尸体现象、法医昆虫学演替规律进行了研究,初步建立了基于高分辨溶解曲线技术(HRM)进行种属鉴定方法,并对相关技术进行了研究。初步建立了基于HRM技术的分子种属鉴定方法,并获得了海南法医昆虫学的相关基础性数据。本研究的开展将为实现法医昆虫学真正运用于法医学实践做出重要贡献。
