Aiming at the urgent need for rapid quantitative detection technology in the field of food safety regulation and to solve the bottleneck problem that it is unable to quantify due to the interference of fruit matrix in SERS detection of pesticide residues, the mechanism of influence of fruit matrix on SERS is systematically studied and the method for overcoming matrix effect is developed. It is planned to conduct research in three aspects as follows: By studying the mechanism of the influence of various components of soluble fruit matrix on the adsorption of pesticides on the surface of SERS substrate, the method via adsorbing pesticides specifically is developed to eliminate the interference of soluble matrix. By studying the scattering and absorption law of incident light and Raman light by different sizes of solid cellulose matrix, the pretreatment method based on membrane filtration is developed to regulate the influence of solid matrix. By establishing a quantitative relationship between SERS intensity and composition and concentration of soluble fruit matrix, concentration and size of solid cellulose matrix, quantitative assessment of SERS intensity fluctuation caused by fruit matrix can be achieved and a theoretical basis is provided for the development of software algorithms that deduct the effect of fruit matrix. The implementation of this project will lay a theoretical foundation for overcoming the matrix effect in SERS detection and provide key technologies for on-site rapid quantitative detection of pesticide residues.
面向食品安全监管领域对快速定量检测技术的迫切需求,针对水果基质所致的表面增强拉曼谱(Surface-enhanced Raman Spectroscopy, SERS)检测农药残留无法定量化这一瓶颈问题,本项目将系统地探究水果基质对SERS的影响机制,并开发克服基质效应的方法。拟在三个方面进行研究:探究可溶性水果基质各组分对农药在SERS基底表面吸附的影响机制,开发农药的特异性吸附方法来克服可溶性基质对吸附的干扰;探究不同尺寸固体纤维素基质对入射激发光和拉曼散射光的散射、吸收规律,开发膜过滤预处理方法来调控固体基质的影响;建立SERS强度与可溶性基质组分和浓度、固体纤维素浓度和尺寸的定量关系式,定量评估水果基质导致的SERS强度波动,为开发扣除水果基质影响的软件算法提供理论依据。本项目的实施,将为克服SERS检测中基质效应奠定理论基础,为农药残留的现场快速定量检测提供关键技术。
本项目以水果中农药残留的SERS定量检测为背景,主要研究了针对拉曼光谱的定量检测方法。在研究中,实现基于深度学习的数据拟合和物质识别方法,为后续农药残留的定量分析打下了分析基础。同时对拉曼光谱技术的推广具有重要意义。
