P16基因在原发性肝癌中的作用及实验治疗研究

In lung cancer therapy and in neuroglial neoplasms therapy, some effective results based on P16 tumor suppressor gene have been gotten. In china, it showed that low level of P16 protein was found in primary hepatocellular carcinomas. Such reasons as gene mutation, deletion and Hypermethylation may cause low level of its product.To make use of P16 gene in primary hepatocellular carcinomas(HCC) therapy, it's necessary to know the status of P16 gene. We collected 32 cases of HCC sample and 32 cases of tissue around the HCC and examined if P16 gene mutation, deletion or hypermethylation exist in these samples by PCR. It showed that 12.5% (4/32) p16 gene deletions and 53.2%(17/32) P16 gene hypermethylation were found in the primary HCCs samples, while no point mutation was identified in the primary HCCs samples. In 32 cases of tissue around the HCC,12.5%(4/32) P16 gene hypermethylation were found,no point mutation and no gene deletion was identified. There was no P16 gene point mutation , no P16 gene deletion and no P16 gene hypermethylation in 8 cases of normal liver tissue.It implied that P16 gene hypermethylation exist in HCCs in china.We examed P16 gene status in 5 hepatoma cell lines and found that P16 gene hypermethylation exist in hepG2.

检测中国大陆地区人原发性肝癌手术标本及肝癌细胞系，了解P16基因缺失、突变或甲基化榭觥８呒谆伟┫赴涤枞ゼ谆恚琍16基因纯合缺失或发生错义突变的肝癌细胞系予P16基因重组腺病毒感染，比较处理前后肝癌细胞系体外生长和体内成瘤性的变化。本项吭て谝庖澹何狿16基因为靶的原发性肝癌的治疗提供实验依据。
