The treatment of acute promyelocytic leukemia (APL) with all-trans-retinoic acid (ATRA) has provided a good model to study anti-cancer differentiation therapy. By using functional genomics approach, such as differential display-PCR (DD-PCR), suppressed subtractive hybrodization (SSH) and cDNA array, we've tried to analyze RA-targeted gene network in order to find out some candidate genes in ATRA-induced differentiation of APL cells. After several years of hard work, a total of 169 ATRA-regulated genes have been identified among which 12 are novel ones. In addition, we've further studied potential functions of those novel genes, including RIG-G, RIG-K and RIG-I.
筛选ATRA调控基因对研究APL发病机理、ATRA诱导分化作用原理和APL耐药机制等有重要意义。目前发现受ATRA调控的基因相当多,但多数为诱导分化的伴随现象,对其调控的关键基因的筛选更具价值。本研究欲应用转染正义和反义cDNA文库的方法筛选ATRA诱导分化相关的关键基因,并克隆再转染进一步验证,找到2-3个起关键作用的ATRA调控基因。
