Poly (amidoamine) (PAMAM) dendrimers with sphere shape (generation 1-5) and fan shape (generation 1-3) were synthesized..The self-assembly ultrathin films with PAMAM dendrimers (carboxylate salts) as polyanions and NDR or DR (Nitrocontaining diazoresin or Diazoresin) as polycations were fabricated and studied. It proves that these unique compounds could recognize and combine with other molecules via electrostatic or H-bond interactions..The peptides of Gly-Asp, Gly-Asp-Gly-Asp, Gly-Asp-(Gly-Asp)2, Gly-Glu and Gly-Glu-(Gly-Glu)2 were prepared and introduced at the periphery of G3.5 PAMAM dendrimers, respectively. These modified dendrimers show bioactivity. They could obviously decrease the reduction rate of FeIII-cyt C by ascorbic acid, especially, the dendrimers with Gly-Asp-(Gly-Asp)2 and Gly-Glu-(Gly-Glu)2 on their surface. Both of the association constants of G3.5-Gly-Asp-(Gly-Asp)2 and G3.5Gly-Glu-(Gly-Glu)2 with cytochrome C are 3.2ⅹ105 and the conformations of them are quite similar to β-sheet by molecular simulation.It has been found that (1) higher generation dendrimers show better affinity with cytochrome C. (2) The peptides with longer chain demonstrate better affinity with cytochrome C. (3) The dendrimers with more modified exterior groups exhibit higher affinity with cytochrome C. (4) Compared with sphere shape dendrimers, the molecules coupled with fan shape dendrimers show stronger affinity with cytochrome C. Dendrimer chemistry and application is an attractive and active study field. It is believed that the application of dendrimers in drug delivery will be a breakthrough. This project is of significance for it establishes the foundation of the study on the applications of dendrimers in biochemistry and medicine..
