The technologies for preparing a drug particle of nano-sized and a suspension containing nano-sized powders have attracted an increasing attention of many researchers, which show a promising prospect in the fields of pharmaceuticals. The purpose of this research is to prepare three kinds of nano-sized drug powders such as cefradine, ibuprofen and salbutamol sulfate by utilized a novel high-gravity technology. In this process a precipitation followed along with a reaction occurred and the particles size and morphology of the powders obtained could be controlled easily. The effect of the process parameters on the properties of the drug powders had been studied respectively under the condition of high-gravity. The results showed that the particle size of cefradine, ibuprofen and salbutamol sulfate prepared was reduced compared with those prepared by traditional process and reached about 200nm, 100~400nm, 500nm and 4mm, respectively. By evaluating the application properties of the drug according to the pharmacopoeia of Chinese (2000eds.), the cefradine particles obtained could meet to the needs of the injection form for the musculature and the suspension for oral. It was understood that the studies of drugs nanosization would have very important significance for enhancing the bioavailability of drugs in the preparations and be helpful to the further research of nano-sized pharmaceutics.
药物颗粒纳米化技术是国际前沿性高技术,具有重大应用价值和医学学术价值。本项目原创性地提出和探索有工业应用前景的超重力沉淀法制备纳米药物颗粒的新方法。研究超重力环境下各物理化学因素对合成颗粒的粒度分布和晶型的影响规律,研究颗粒粒度与药物释放性能及生物功能之间的关系,建立制备工艺—颗粒粒度形貌与结构—药物性能链的关系模型。
