Tulip is one of the most important ornamental crops in the world. It is widely used as cut flower, pot plant and garden flower. Tulip breaking caused by Tulip Breaking Virus (TBV) is the most important and severe virus disease for tulip. It significantly lower the quality of cut flower and reduce bulb production. With the rapid expansion of tulip cultivation area, TBV is spreading worldwide. It is an effective method to control virus diseases by selection of disease resistant cultivars. This study is going to evaluate the TBV resistance of breeding populations, common cultivars in China as well as some wild tulip species in China. Find out TBV resistant materials is the basis of breeding disease resistant cultivars. Tulip has a long juvenile stage, which means selection of new cultivar through conventional breeding approach is a time consuming process. Breeding efficiency is very low. Marker assissted selection (MAS) benefits fast screening of individuals in populations, thus accelerate the breeding process. This study is going to enrich the genetic maps which were constructed in our former study by adding more SNP and SSR markers, and integrate the parental maps. The TBV resistance gene will be located in the genetic map through linkage analysis. Molecular markers which closely linked to the TBV resistance gene is going to be identified and verified. Result of this study will provide theoretical basis for marker assissted selection of TBV resistant tulips.
郁金香是世界上重要的观赏植物之一,广泛应用于切花、盆栽和庭院种植。碎色花瓣病由郁金香碎色病毒(TBV)引起,严重影响郁金香切花品质、降低种球质量和产量。随着近年郁金香栽培规模的迅速扩大,TBV在世界范围内呈蔓延趋势。通过选育抗病新品种来防治病毒病的发生是一种行之有效的方法。本项目对国内常用郁金香品种、我国郁金香野生资源及本课题组获得的部分杂交群体的TBV抗性进行鉴定,筛选抗病种质,为抗病杂交育种提供基础。因郁金香幼年期很长,以传统的杂交育种方式选育新品种耗时长、育种效率低。采用分子标记辅助选择有利于对后代进行快速筛查,加速育种过程。本项目在前期构建的遗传图谱基础上,增加SNP和SSR标记,提高图谱饱和度,获得整合图谱,然后通过连锁分析将郁金香碎色花瓣病抗性基因定位于遗传图谱上,筛选与抗病基因连锁的分子标记并进行验证,为今后郁金香抗碎色花瓣病分子标记辅助选择育种提供理论基础。
