For insect overwintering as an anti-cold strategy in a changing climate,its somatic cells are modified to overexpress a variety of proteins, in particular, cold induced proteins (CIPs). There is a continental climate with much long and cold winter in Changbaishan Mount area; natural and chronic exposure of maintained honeybee (Apis mellifera) species to this rigorous winter contributes its protective cold-resistant property. To quantitatively determine the pattern of CIPs in honeybee and thereby reflect its cold-resistant property, in the research, a proteomic analysis of experimental honeybee is to be at first carried out to identify protein candidates of CIPs, and furthermore, both immunological analysis and quantitative transcript analysis are to be carried out to validate them. It is suggest that the observed protein bioinformatics datasets might characterize cold-resistant property of honeybee, the analysis of protein bioinformatics might provide with relevant aspects of cold-resistant mechanism of honeybee,and the compositional pattern of CIPs in honeybee might be biological index for whether freeze-avoiding insect or insect with freeze tolerance. Original and Novel: After natural and chronic exposure of maintained honeybee species to this rigorous winter is found to contribute its protective cold-resistant property, an analysis of protein bioinformatics, including proteomic, immunological and transcriptomic analyses, is to be carried out to reveal cold-resistant mechanism of honeybee.
昆虫在抵御寒冷过程中它的体细胞异常表达冷诱导蛋白质(CIPs)。长白山漫长的冬季月份和寒冷的冬季温度为低温驯化西方蜜蜂的抗寒性提供了天然的冷暴露处理。为了定量测定西方蜜蜂CIPs,和以此定量测定值为指标反映西方蜜蜂的抗寒性,本研究首先通过蛋白质组学分析,在蛋白质组水平鉴定CIPs的候选蛋白质,然后通过免疫学分析和转录物核酸定量分析,分别在蛋白质翻译、转录水平证明CIPs。这提示西方蜜蜂的CIPs蛋白质生物信息学数据能够成为表征西方蜜蜂抗寒性的生物学指标,西方蜜蜂的CIPs蛋白质生物信息学分析能够揭示了西方蜜蜂的抗寒机制,西方蜜蜂的CIPs构成成分能够判定西方蜜蜂属于耐受结冰昆虫或避免结冰昆虫。特色和创新:从长白山气候特征中发现了冷暴露对西方蜜蜂抗寒性的影响,并且通过CIPs蛋白质组学分析、免疫学分析和转录物核酸定量分析,揭示西方蜜蜂的抗寒机制。
