The hydrogels as new biomedical materials have attracted much attention. Among them, the self-healing hydrogel as novel injectable hydrogel has high research value. Our research group has developed a chitosan based self-healing hydrogel system. This hydrogel has the advantages of simple preparation using cheap raw materials. The hydrogel also has good biocompatibility and responds to many external stimulants. In this project, based on our previous studies, we will further investigate the basic theory and explore the biological and medical applications of the chitosan based self-healing hydrogel. In the preliminary experiments, the chitosan based self-healing hydrogel, used as injectable hydrogel, showed good results in the clinic fields for the drug delivery and wound healing, demonstrating the feasibility of the project. In this project, we will further develop this hydrogel system, design the polymer components of the hydrogel, optimize the preparation of the self-healing hydrogel, study the structure-property relationship, and establish the solid chemical foundation for its biomedical applications. We will also systematically study this hydrogel system at multi-levels, through chemical syntheses, animal experiments and clinical tests, to demonstrate the irreplaceability of the self-healing property of the hydrogels as drug carriers. We expect that our research can provide a theoretical and experimental foundation for the practical applications of the chitosan-based self-healing hydrogels.
水凝胶作为一种新的生物医用材料备受关注。作为新型可注射型水凝胶,自愈性水凝胶更具有重要的基础与应用研究价值。本课题组研制出了一种基于壳聚糖的自愈性水凝胶体系,该体系原料价廉易得、制作工艺简单、具有良好的生物相容性以及多重生物刺激响应性。以之前的研究工作为基础,我们将在本项目中进一步深入研究壳聚糖基自愈性水凝胶的基础理论并发掘其生物医用价值。初步实验显示,壳聚糖基自愈性水凝胶作为可注射型水凝胶,在药物缓释放和促进创伤愈合两方面均取得了较好的效果,表明了本项目的可行性。在本项目的实施中,我们将通过高分子成胶组分的设计,优化可注射型自愈性水凝胶的制备,研究材料结构与性能的关系,为其生物医学应用奠定坚实的化学基础。再通过细胞、动物与临床实验,从多角度、多层次进行深入的生物学研究,验证水凝胶的自愈性在其作为药物载体时可起到不可替代的作用,为壳聚糖基自愈性水凝胶的最终实际应用提供相关的理论及实验基础。
本项目以制备具有临床应用的生物水凝胶为导向,在性能调控和生物应用两方面进行研究,取得的主要成果有:.1)可注射型自愈性水凝胶的制备与性能研究:以天然产物壳聚糖为原料,利用动态化学形成具有良好的生物安全性的自愈性、自适性的水凝胶。作为细胞载体时,该水凝胶对充分发挥药效、提高治疗效果起到关键作用。.2)可注射型自愈性水凝胶用于药物缓释放和肿瘤治疗的研究:自愈性使该水凝胶在注射入体内后可以修复为一个整体,为发挥药效提供了必要条件。作为可注射型药物载体时,表现出良好的肿瘤治疗效果。.3)可注射型自愈性水凝胶促进创伤愈合的系统研究:该体系秉承了水凝胶的自愈性,壳聚糖的抗菌特性,也具备了药物缓释放的性质,在动物实验和初期临床实验中体现出较好的愈合能力,在生物医学、临床应用方面具有较大潜力。.自2015年以来,已发表SCI论文70余篇,申请专利1项,培养博士6名,博士后2名,访问教授2名,多次参加国际会议并做大会或邀请报告。
