Uveal melanoma (UM) is the most common primary intraocular malignant tumor in adults. Radiotherapy is the preferred treatment for most cases of UM. However,some UM cells are relatively radioresistant.Although microRNA (miRNA) have recently been recognized as important regulators of gene expression and play an important role in response to cellular stress including radiation, it is not well examed if miRNA play any roles in UM cells response to radiation.Therefore,we investigated whether any changes in the expression profiles of miRNA in UM cells.Our current result suggested expression of miRNA in let-7 family significently changed after radiation.We raise the hypothesis:let-7 might play an important role in UM cells response to radiation through regulated the key protein of cell cycle,such as Ras、C-myc.In this project we will try to discover the mechanism of let-7 affected the radiosensitivity of UM cells by in vivo and in vitro study.We believe this project will bring new bright on the radiotherapy for UM.
葡萄膜黑色素瘤(Uveal Melanoma,UM)是成人最常见的眼内恶性肿瘤。放疗是其主要治疗方式,但部分UM对放射不敏感。研究表明miRNA表达变化在放射引起的细胞应激反应中发挥作用,进而影响细胞放射敏感性。迄今对UM放射敏感相关miRNA仍知之甚少。我们的预实验结果提示,放射处理后UM细胞中let-7家族miRNA表达显著改变,但其对UM放疗敏感性的具体影响和作用机制还有待进一步探讨。为此,我们提出假说:let-7可能通过影响Ras、C-myc等细胞周期调控蛋白表达,导致UM细胞放射敏感性的改变。为了验证这一假说,我们将通过人UM的 OM431、OCM1等细胞系,采用Real-time PCR,Western blot,慢病毒载体转染等手段,从分子、细胞、动物整体水平等多方面探讨let-7在UM放射敏感性中的作用,明确其调控机制,从而为提高UM放射疗效提供新思路。
葡萄膜黑色素瘤(Uveal Melanoma,UM)是成人最常见的眼内恶性肿瘤。放疗是其主要治疗方式,但部分UM 对放射不敏感。研究表明miRNA 表达变化在放射引起的细胞应激反应中发挥作用,进而影响细胞放射敏感性。迄今对UM 放射敏感相关miRNA 仍知之甚少。我们通过芯片检测发现放射后UM 细胞中let-7 家族miRNA 表达显著改变。体外实验表明过表达 let-7b可以使肿瘤细胞活力明显降低,增强UM细胞的放射敏感性,同时发现let-7b可能是通过影响Cyclin D1的表达,进而改变肿瘤细胞的细胞周期,从而起到调控UM细胞放射敏感性的作用。动物实验提示 let-7b可以在体内增强UM细胞的放射敏感性,进而增强放射对UM的治疗效果。本研究中,我们通过OM431、OCM1 等细胞系,采用Real-time PCR,Western blot,慢病毒载体转染等手段,从分子、细胞、动物整体水平等多方面研究了let-7b 在UM 放射敏感性中的作用,探讨了其调控机制,从而为提高UM 放射疗效提供新思路和作用靶点。
