Oxidative stress can lead to hepatocytes injury, which is the most important pathogenic mechanism in acute or chronic hepatic diseases. HO-1 reveals protective effect in deal with oxidative stress. Maslinic acid(MA) exhibits antioxidant effect, but its exact antioxidant mechanism through regulating HO-1 remains unclear. In our project H2O2-induced the primary hepatocytes of mice will be used as hepatocytes injury models. The effect on hepatocyte inury will be respectively observed after MA administration. Some indexes will be detected by biochemical methods, in order to identify its antioxidant effect. In addition, the antioxidant effect of MA will be focused so as to identify the activity, mRNA and protein of HO-1. The target molecule of MA will be determined and its relative molecular mechanism will be explored. The present study will provide useful evidences for the antioxidant effect of MA and its exploitation.
氧化应激能导致肝细胞损伤,成为各种急性或慢性肝脏疾病的重要致病机制。HO-1是对多种氧化应激有保护作用的蛋白。山楂酸具有较好的抗氧化效果,是否能够通过调控HO-1发挥抗氧化功能尚不清楚。本项目拟以H2O2诱导的小鼠原代肝细胞作为肝细胞氧化损伤模型,观察给予MA后,对肝细胞氧化损伤情况的影响,并采用生化方法检测相应的指标变化,以确定MA抗氧化的作用,同时测定HO-1的酶活性、mRNA和蛋白水平的变化,确定MA作用的靶分子,探讨其抗氧化的细胞分子机制,为MA抗氧化功能的开发提供理论依据。
