By monitoring the biodegradable organic matter and the volatile fatty acid (VFA) content of the refuse, the VFA concentration, pH and B/C of the leachate, the production rate and composition of the landfill gas, the effect of the acid neutralizing intermediate layer on the VFA accumulation in the landfill is to be investigated. The effect of the acid neutralizing intermediate layer on the stabilization and substance metabolism of the landfill can also be revealed. Besides, the effect of the acid neutralizing intermediate layer on the distribution pattern of hydrolytic bacteria, hydrogen-producing acetogenic bacteria and methanogenic bacteria is to be studied by using the high-throughput sequencing techniques and bioinformatics methods. The quorum sensing among the bacteria flora is to be explored by analyzing the release of auto-inducers. Based on the above results, the effect of quorum sensing on substance metabolism-community dynamics and substance metabolism-functional gene expression can be clarified, which can help understand the influence mechanism of the acid neutralizing intermediate layer on the stabilization of the landfill. The result of the project can enrich the method of the fast stabilization of the landfill. Furthermore, it can reveal the influencing mechanism at the molecular ecology and functional gene expression level.
以含酸缓冲型中间覆盖层的填埋场反应器为模型系统,通过监测垃圾体可降解物、挥发性脂肪酸(VFA),渗滤液VFA、pH、B/C,填埋气产量和组分等指标,研究酸缓冲型中间覆盖层对填埋场VFA积累的缓释作用,揭示其对填埋场稳定化进程和物质代谢的影响;采用高通量测序和生物信息学方法探索酸缓冲型覆盖层对填埋场中水解菌群、产氢产乙酸菌群和产甲烷菌群分布规律的影响;以群体感应为切入点,解析群体感应信号分子、菌群变化、功能基因表达和物质代谢之间的关系,探究酸缓冲型中间覆盖层对水解菌群、产氢产乙酸菌群和产甲烷菌群协同作用关系的影响,从而揭示酸缓冲型中间覆盖层加速填埋场稳定化的分子生态机理。项目的预期研究成果将丰富实现填埋场快速稳定化的技术手段,并从微生物生态、功能基因表达等层面揭示酸缓冲型中间覆盖层加速填埋场稳定化的机理。
项目执行期间,搭建了含酸缓冲型中间覆盖层的填埋场反应器,通过监测填埋气的产量与组分(CH4和CO2),渗滤液的理化性质(pH、VFA、盐度、TOC),垃圾体的降解,研究了酸缓冲型中间覆盖层对填埋场VFA积累的缓释作用,揭示了其对填埋场稳定化和物质代谢的影响。采用了分子生物学和生物信息方法,研究了酸缓冲型覆盖层对填埋场微生物群落分布的影响,探究酸缓冲型中间覆盖层对水解菌群、产氢产乙酸菌群和产甲烷菌群协同作用关系的影响,从而揭示了酸缓冲型中间覆盖层加速填埋场稳定化的分子生态机理。项目研究成果在Chemical engineering journal、Ecological engineering、Environmental science and pollution research、RSC advance等期刊发表SCI论文5篇,授权发明专利2项,实用新型专利1项,申请发明专利和实用新型专利各1项,并建立了较为广泛的国际学术交流与合作关系。项目执行期间项目负责人在2017年入选浙江省高等学校中青年学科带头人,2018年入选台州市211人才第一层次培养人员。
