Object:To observe the effect of γ-ray irradiation on rat vascular smooth muscle cells(VSMCs)and investigate the mechanism of inhibitory effect for γ-ray irradiation on VSMCs. Methods:Dose-survival curve of VSMCs was figured by colony formation. The effect of γ-ray irradiation on the proliferation of VSMCs was observed by 3H incorporation. Flow cytometry and "DNA Ladder " was used to detect the apopotogenic effect and cell cycle change of γray irradiation on VSMCs. "Western Blot" was used to investigate the expression of Tp53,cyclinD and PCNA after γray irradiation. .Results :The values of D0,Dq,D37 and N for VSMCs were 1.95Gy, 1.76Gy, 3.71Gy and 2.47 respectively. The inhibitory effect of γ-ray irradiation on VSMCs proliferation was dose-dependent, being stronger along with increase of dose. VSMCs did not undergo apoptosis within 48 hours after γ-ray irradiation. VSMCs was arrestted in G1 phage afterγray irradiation. The expression of p53 was increased afterγ-ray irradiation, while the expression of cyclinD and PCNA was decreased.Conclusion:γ-ray irradiation could inhibit the proliferation of VSMCs , the main mechnism of which is the cell cycle arrest, killing effect and inhibition of mitosis of VSMCs after irradiation. p53、cyclinD and PCNA played an important role in this course. γ-ray irradiation could not induced VSMCs in vivo apoptosis .
