The lung is a serious highest incidence dysfunction and the earliest occurrence organs after inhalation injury, Mesenchymal stem cells have a strong ability to endocrine and immune function, it is focus to transplant stem cells in tissue repaired research, but directly transplanted MSCs having the tumorigenic, embolism, abnormal differentiation, immune regulation and other non-controllable risks, exosomes from mesenchymal stem cells can be delivered to the secretion system of extracellular matrix membrane when stem cell resting or activation state, exosomes contain not only mesenchymal stem cell active substance and the selective assembly, targeting delivery and efficient repair damaged tissue, safe, easy to store chemical stability,and so on.It is an important field of stem cell research.In our study,collecting MSCs exosomes after overexpressing JAM-A gene, injecting exosomes by intravenous method to smoke inhalation injury model,we found that it could inhibit inflammation and the promote pulmonary vascular repairing, and further explored by cell experiments of lung injury repair mechanism. Exosomes has been found that it not only has a biological characteristic,but also MSCs can carry genetically modified active substance, it will offer a good reference method targeted therapy to acute lung injury.
肺脏是严重吸入性损伤后功能不全发生率最高和发生最早的器官,间充质干细胞具备较强的内分泌能力和免疫调节功能,移植干细胞进行组织修复是研究热点,但直接移植MSCs具有致瘤、栓塞、异常分化、免疫调节不可控等风险,间充质干细胞外泌体(exosomes)能够在干细胞静息或活化状态下释放到细胞外基质中的膜分泌体系中,外泌体不仅含有间充质干细胞的活性物质,而且具有选择性组装、靶向性投递、高效修复受损组织、安全性高,化学性质稳定易于保存等优点,是干细胞领域具有重要研究价值的新兴热点。本研究过表达的JAM-A基因后收集MSCs外泌体,通过静脉注射方法在烟雾吸入性损伤模型上进行研究,发现能够很好的抑制炎症反应和促进肺部血管修复,通过细胞实验进一步探讨修复肺损伤机制,为外泌体开发成为一种既具有MSCs生物学特性又能携带基因修饰的活性物质,靶向治疗急性肺损伤提供方法借鉴和参考。
