课题组通过国家自然基金项目“太阳病(风寒表证)‘欲解时’与白介素变化的相关性研究”及“太阴病(脾阳虚证)‘欲解时’与环磷酸腺苷的相关性以及理中汤干预研究”的课题,初步研究了人体免疫功能与太阳病发生、发展规律的内在联系,探讨了太阴病的病理生理基础及欲解时的意义。在此基础上,本课题拟进行“少阴病(心肾阳虚证)“欲解时”与实验大鼠心肌组织PGC-1α能量信号通路变化及四逆汤干预研究”,通过实验动物心肌组织PGC-1α、NRF-1、mtTFA,及ATP、PCr、CK、BNP等指标的变化,探讨少阴病(心肾阳虚证)的生理病理学基础;并通过四逆汤干预的效果研究,探明少阴病(心肾阳虚证)“欲解时”的时间治疗学意义,为全面而深入地阐释六经“欲解时”的科学性及客观性提供实验依据。
课题组通过国自然项目“太阳病风寒表证‘欲解时’与白介素节律变化的相关性研究”及“太阴病脾阳虚证‘欲解时’与环磷酸腺苷的相关性以及理中汤干预研究”课题,初步研究了人体免疫节律与太阳病发生、发展规律的内在联系,探讨了太阴病的生理病理基础以及欲解时的时间治疗学意义。在此基础上,为完整认识六经病“欲解时”的本质,本课题拟通过对少阴病“欲解时”心肾阳虚证实验大鼠造模前后心肌组织能量代谢通路关键蛋白PGC-1α、NRF-1、mtTFA及心肌酶CK、CK-MB、LDH、AST、甲状腺素 T3、T4及TSH指标在少阴病欲解时中心时丑时(1:00-3:00)及其对时未时(后称非欲解时13:00-15:00)表达的差异和变化规律的研究,探讨少阴病心肾阳虚证“欲解时”的生理病理基础;并通过四逆汤干预效果的研究,探讨少阴病心肾阳虚证“欲解时”的时间治疗学意义,为全面阐释六经“欲解时”的科学性以及客观性提供思路、方法及实验依据。. 结果发现正常组、模型组、药物组(四逆汤)大部分时间段PGC-1α、NRF-1、mtTFA蛋白、CK、CK-MB、LDH、AST、T3、T4、TSH各指标含量均为欲解时高于非欲解时;三组PGC-1α、NRF-1、mtTFA、LDH、AST、T3、T4、TSH指标基本同步变化,节律一致,且显示了一定的昼夜节律; NRF-1、mtTFA蛋白含量、CK、CK-MB、LDH、AST、T3、T4含量均为模型组高于正常组。. 实验结果说明实验大鼠心肾阳虚证病变存在着心肌组织能量代谢障碍、心肌损伤、甲状腺功能紊乱。经四逆汤干预后,各指标均有改善,说明四逆汤对心肾阳虚证大鼠有保护心肌组织、纠正甲状腺功能、改善心肌组织能量代谢的效用。亦验证了少阴病欲解时时辰节律客观性、及“欲解时”的另一层含义“欲剧时”。
