Embryonic exposure to addictive drugs have serious harm on offspring's physical and mental health, especiallly changing offspring's susceptibility to addictive drugs. The study will use chicks as a model animal to study the critical period of embryonic morphine exposure on addictive susceptibility and the involved role of BDNF. This study will concern on avian caudolateral nidopallium (NLC), the presumed functional analog of the mammalian prefrontal cortex. We will utilize increased morphine-induced psychomotor activity by prenal morphine exposure as animal model: 1) using immunohistochemistry and calcium imaging method, we will analysis the impact embryonic morphine exposure on the number of neurons and neuronal activity in NCL; 2) using PCR and ELISA method, we will further analysis the impact of embryonic morphine exposure on BDNF mRNA and protein levels in NCL; 3) by administration exogenous BDNF, we will examine the possible inversed effect of exogenous BDNF on prenatal morphine exposure induced changes in behavioral, number of neurons and neuronal activity in NCL. This presenst investigation not only advance our understanding of the neural mechanisms of embryonic addiction drug exposure on offspring's behavior but also provide basis research evidence for explore an effective treatment to prenatal drug exposure.
胚胎期成瘾药物暴露严重危害子代身心健康,改变成瘾易感性。本项目以小鸡为模式动物,研究胚胎期吗啡暴露影响子代成瘾易感性的关键期以及脑源性神经生长因子(brain derived neurotrophin factor, BDNF)的作用。本项目关注鸟类的caudolateral nidopallium(NLC)核团,其功能与哺乳动物的前额叶皮层类似。研究以胚胎期吗啡暴露增强子代吗啡诱导的精神运动兴奋性为模型:1)采用免疫组化和钙离子成像的方法,分析胚胎期吗啡暴露对NCL的神经元数量及活动性的影响;2)使用PCR和ELISA方法,分析胚胎期吗啡暴露对NCL的BDNF的mRNA和蛋白的影响;3)通过外源性补充BDNF,考察BDNF对胚胎期吗啡暴露所致成瘾易感性、神经元数量、神经元活动性改变的翻转作用。本项目为深入理解胚胎期成瘾药物暴露影响子代行为的神经机制以及探索有效的治疗方法提供基础研究证据。
