Hepatitis B is an epidemic disease that seriously endangering human health. China, a high incidence area of hepatitis B, is in urgent need for safe and effective anti-hepatitis B drugs. However, at the present stage, the main treatment of hepatitis B is to take preventive measures like vaccination, and there are few drugs which have exact effect of protecting the liver and resisting the virus.It has been found in recent years' researches that many Genus Wedilia plants have significant anti-hepatitis activity, for they contain Wedelolactone that has a protective effect on the liver injury.Wedelia prostrata, one of Genus Wedilia plants, which is a rich resource on both sides of the Taiwan Strait, has been widely applied in the folk medicine for the treatment of hepatobiliary diseases. However, few research has been done on its chemical constituents and pharmacological effects. Therefore, this project is intended to systematically study the chemical constituents of Wedelia prostrata that grows in Fujian province by adopting the activity-tracking measure, making use of various advanced chromatographic separation technologies and structural identification means measures, taking anti-hepatitis B activity as an indicator. It is expected to discover new anti-hepatitis B substances, and to clarify their effective substance for the treatment of hepatitis B.Based on clinical experience of traditional Chinese Medicine and abundant resources of the Chinese herbal medicine in our country, the project aims to find high-efficient and low-toxic anti-hepatitis B substances from Chinese folk medicine. It has very important significance to the development and application of Chinese folk medicine and the modernization of Chinese tradicional Medicine.
乙型肝炎(乙肝)是一种严重危害人类健康的流行性疾病,我国为乙肝的高发区,迫切需要安全有效的抗乙肝病毒的药物。现阶段,乙肝的治疗以接种疫苗等预防手段为主,具有确切的保护肝脏、抗乙肝病毒疗效的药物较少。蟛蜞菊属的多种植物在近年的研究中均被发现具有显著的保肝抗炎活性,其中蟛蜞菊内酯对肝损伤有明显的保护作用。海峡两岸资源丰富的植物卤地菊同样隶属于蟛蜞菊属,该植物在民间也广泛的运用于肝胆疾病的治疗,但是关于其化学及药理学研究的报道均较少,因此本项目拟采用活性追踪的方法,以抗乙肝病毒活性为指标,利用先进的色谱分离技术和结构鉴定手段,对闽产卤地菊的化学成分进行深入系统的研究,以期发现结构新颖的抗乙肝病毒活性成分,阐明其用于治疗乙肝的物质基础。本项目立足于传统中医药的临床经验和我国丰富的中草药资源,致力于从民间药物中寻找和发现高效低毒的抗乙肝病毒活性成分,对民间药物的开发运用以及中药现代化都具有重要意义。
卤地菊(Wedelia prostrata(Hook. et Arn.)Hemsl.)为菊科蟛蜞菊属植物。对蟛蜞菊属植物研究调查表明,其主要含有二萜,倍半萜,黄酮和有机酸等化学成分。二萜尤其是对映-贝壳杉二萜类是蟛蜞菊属植物的主要活性成分,具有抗菌、抗病毒、抗氧化、抗肿瘤等生物活性。. 本项目采用活性追踪的方法,以抗肿瘤活性为指标,利用先进的色谱分离技术和结构鉴定手段,对闽产卤地菊的化学成分进行深入系统的研究,阐明其抗肿瘤的物质基础。本项目用石油醚、乙酸乙酯、正丁醇三种不同极性的溶剂依次对卤地菊的乙醇提取物进行萃取,得到三个萃取部位。采用硅胶柱层析、聚酰胺柱层析、葡聚糖凝胶柱层析、大孔吸附树脂柱层析和半制备型高效液相色谱等分离技术对石油醚部位浸膏进行分离纯化。利用现代波谱技术(HR-ESIMS,1H NMR,13C NMR,DEPT,2D NMR等)对分离得到的化合物进行结构鉴定,实验分离鉴定得到了18个化合物。包括1个单萜,2个倍半萜,8个二萜,2个三萜,1个甾醇,1个黄酮,3个有机酸。其中对映-贝壳杉-3α-苯丙酰基-19-酸(1)是新化合物,另有5个化合物为首次从卤地菊植物中分离得到。同时,参考萜类化合物的HSCCC溶剂体系,结合TLC和HPLC分析以及测定分配系数,筛选出正己烷/乙酸乙酯/甲醇/水体系作为溶剂系统,并对溶剂系统和流速进行优化,成功应用HSCCC对卤地菊的石油醚部位化学成分快速分离并得到了6种萜类单体化合物。体外抗肿瘤研究表明,从卤地菊中分离所得的某些特定的化合物对不同的肿瘤细胞有较强的抑制作用,并对不同的肿瘤细胞有一定的选择性,表明卤地菊抗肿瘤活性方面具有一定的潜力和研究价值。. 综上所述,本项目通过三年研究完成了预定的目标,所获得的项目成果对民间药物的开发运用以及中药现代化都具有重要意义。
