Hemorrhagic fever with renal syndrome (HFRS) is known to cause serious and often fatal human diseases, but the pathogenesis of it are still unclear. It is known that the damage of a number of tissues and cells of the body, especially the capillary endothelia cells, by hantavirus is primary and major mechanism of HFRS occurring and progressing. The research showed that abnormal immune modulation function played important role in HFRS occurring. To explore the effect of immune system to the pathogenesis of HFRS, the endothelial cells and human lymphocytes were infected by hantaan virus, and RT-PCR, immunofluorescence assay and Confocal Laser Microscope etc. were used to detected the expression of some cytokines and transcription factors in the infected cells. The results showed that the virus infection could induce different cytokines in human endothelial cells and human monocytes. In human endothelial cells, it could induce the expression of ICAM-1 and eNOS. However, it could induce the expression of IL-6 in human monocytes. Meanwhile, it is showed that hataan virus infected human monocytes could also result in the activation of IRF-3/IRF-7 , the expression of beta chemokine RANTES and the raise of Ca++ in cells. These results lays the foundation for further research on pathogenesis of HFRS.
