Root caries is a common disease of oral medicine, due to high prevalence, high resistance rates,and low detection rates of cases in china.Dental plaque is a complex bacteria group of oral biofilm structure. The chinese herbal formula of galla chinensis and Cimicifuga foetida received much attention of multiple domestic and foreign scholars. However, the mechanism is unclear. Representatives of early following school found in clinical practice, the effect of prescription main of galla chinensis and cimicifuga foetida is significant and exact, but its pharmacological effects and mechanism of action is unclear, so clinical application of the method is affected. We have isolated Predominant pathogenic strains from root caries ----Streptococcus mutans and actinomycetes in preliminary study, and studied the antibacterial and bactericidal effect of the extract of galla chinensis, showing the value for further research. In this study,we intend to establish model of predominant strain, based on activity tracking method of HPLC-based activity profiling, select active substances of anti-root caries from galla chinensis and cimicifuga foetida , and learn its role characteristics, through validation studies of rantibacterial activity of the active ingredients, further understand its mechanism, expecte to find anti-root caries medicine which Structure and mechanism of action is clear, to provide scientific reference for further clinical applications and new clinical ideas for galla chinensis and Cimicifuga foetidaanti-gallic Cimicifuga to resist root caries.
根面龋(RSC)是口内科常见病,具高患病率、高耐药率及病例发现率低等特点。牙菌斑是一种具有复杂细菌群结构的口腔生物膜。中草药防治RSC由于不易产生耐药性,而倍受国内外关注。前期,我们在挖掘古籍文献基础上并结合现代临床实践发现中药五倍子配伍升麻的"药对"治疗RSC疗效显著确切,且效果优于五倍子单用,提示中药抗RSC具有明显优势;进而研究了五倍子提取物对RSC优势致病菌的抑制和杀伤作用,但"药对"配伍后呈现的优效性其协同作用及其机制尚不清楚,其有效物质也不清楚,从而制约了这一有效方剂在临床上的推广应用。本项目拟以RSC优势菌株为模型,采用HPLC-based activity profiling活性跟踪方法,筛选升麻和五倍子中抗RSC活性物质,并借鉴正交试验研究有效组份之间对RSC细菌生物膜的作用及其机制,有望从中发现作用和机制相对明确的抗RSC药物有效组份,从而为其临床推广应用提供重要基础。
