Altered metabolism is a key feature for tumor cells. Understanding these unusual metabolic pathways may open doors to new therapeutic strategies against tumor. While metabolic defects have been investigated extensively in differentiated tumor cells, much less attention has been directed to the metabolic properties of cancer stem cells (CSCs). Using a mechanical method to select and grow CSCs by culturing single tumor cells in three-dimensional (3D) soft fibrin gels, the applicant surprisingly found that liver CSCs reprogram glucose metabolism by hijacking and upregulating phosphoenolpyruvate carboxykinase (PEPCK). However, upregulated PEPCK in CSCs didn’t mediate gluconeogenesis due to the downregulation of FBP1 and G6Pase expression in those CSCs. Considering the important side-branches in gluconeogenesis including the serine and glycerol-3-phosphate pathways, this study hypothesize that PEPCK promotes glucose side-branch metabolism so to quickly provide serine, glycerol-3-phosphate and other building blocks for CSC growth. Together, this study will clarify the role of PEPCK in side-branches of gluconeogenesis in hepatocarcinoma stem cells, leading to potential therapeutic target against hepatocarcinoma.
代谢异常是肿瘤细胞的重要生物学特征,解析肿瘤代谢过程不但有助于阐明肿瘤发生的内在变化机理,而且有望为临床肿瘤治疗提供新的策略。尽管目前肿瘤代谢研究方兴未艾,但缺乏对肿瘤干细胞代谢特征的探究。申请人利用课题组前期建立的三维纤维蛋白软胶体外筛选肿瘤干细胞的技术,对获得的肝癌干细胞进行了研究,发现其上调表达糖异生的限速酶PEPCK,却不表达另外两个限速酶FBP1和G6Pase,表明肝癌干细胞尽管高表达PEPCK,但却不能完成糖异生。由于糖异生途径中存在不同的侧枝代谢去向,可生成丝氨酸或3-磷酸甘油。据此,本项目提出假设:PEPCK促进肝癌干细胞糖异生侧枝代谢途径的进行,进而促进丝氨酸、3-磷酸甘油等中间代谢产物的生成,以满足其自身旺盛的代谢需求。本项目的开展将阐明PEPCK在肝癌干细胞糖异生侧枝代谢途径中的作用,并有望从肝癌干细胞代谢的角度为肝癌治疗提供新策略。
肿瘤相关巨噬细胞是肿瘤治疗的潜在靶点,但其来源和功能并不清楚。本部分研究发现:肿瘤细胞来源的微颗粒通过激活cGAS/STING/TBK1/STAT6信号通路诱导巨噬细胞向M2型肿瘤相关巨噬细胞极化,进而促进肿瘤的生长、迁移及肿瘤干细胞的发展。本部分研究阐述了肿瘤细胞来源的微颗粒在肿瘤微环境重塑中的重要作用,有助于理解肿瘤和巨噬细胞的信息交流。.CD8+ 记忆性 T 细胞在抗感染和抗肿瘤免疫中发挥重要作用。代谢调控在维持记忆性T细胞稳定中至关重要,但是代谢信号调控记忆性T细胞的形成和维持的分子机制仍不清楚。本部分研究发现:调控糖酵解,三羧酸循环的糖异生限速酶Pck1在CD8+ 记忆性 T 细胞内的表达上调,进而促进糖原合成,糖原进而分解生成葡萄糖-6-磷酸,并通过磷酸戊糖途径生成大量的NADPH,从而确保记忆性T细胞内高水平的还原型谷胱甘肽(GSH)。破坏“Pck1-糖原代谢-磷酸戊糖途径”导致GSH/GSSG比例下降和活性氧(ROS)水平增加,从而抑制CD8+ 记忆性 T 细胞形成和维持。这一发现对于发展高效的T细胞免疫疗法提供了重要的代谢理论基础。
