Current interest in the development of noninvasive molecular imaging is being shifted toward multimodality imaging, as no single imaging modality possesses all of the ideal traits of being quantitative and provides both high resolution and sensitivity. The combination of non-ionizing magnetic resonance imaging (MRI) and optical techniques has attracted attention because of their highly complementary capabilities for anatomical resolution and detection sensitivity. One of the key technologies is to design and synthesis molecular probes which have good biological compatibility and specific target characters. Silicon quantum dots (SiQDs) are an attractive platform for building multimodality imaging probes, and it tends to require short-wavelength excitations which are subject to tissue scattering and autofluorescence artifacts. Furthermore, SiQDs are expected to be an ideal candidate in many biological applications because of their proven biocompatibility Compared with other methods, 19F-MRI technique can detect contemporarily and quantificationally,it also have high imaging contrast and sensitivity. This proposal is intended to create one of multifunctional molecular probes based on SiQDs and 19F, and apply such probe with high sensitive optical and MRI properties to the human pulmonary adenocarcinoma cell (A549 cell).
肺癌是我国目前死亡率第一的恶性肿瘤,特别是近年来随着环境污染问题的加剧,其发病率逐年递增。早期诊断是扑灭癌症的关键之一,因此发展更为全面和精密的诊断技术受到人们的广泛关注。现有的磁共振和光学成像技术是目前癌症诊断的重要工具,但是单一技术所提供的信息有限。本项目拟针对肺癌,发展磁共振/荧光双功能成像技术,开发新的高灵敏和高特异性肺癌检测的分子探针,并将其应用于人肺腺癌A549细胞的分子影像学研究中。该探针拟以具高灵敏度和高成像对比度的19F作为磁共振检测基元,以生物兼容性和光学活性良好的硅量子点作为荧光检测基元,以实现对癌细胞A549的高灵敏成像和特异性识别,为肿瘤的早期诊断提供帮助。
肺癌是威胁人类健康的重要疾病,其发病率和死亡率逐年上升,因此,发展多模态分子影像技术以获取多方位,高分辨信息,明确肺癌的分子机制并为其早期诊疗提供帮助,具有重要的理论意义和实用价值。本项目设计并构建一系列适用于磁共振和荧光分子影像的双功能19F MRI/荧光分子探针,并使得制备的探针具有高特异性的靶向能力,实现对人肺腺癌A549 细胞的高灵敏、特异性检测,在细胞和分子水平上实现了实时的动态跟踪和在体成像,显示分子探针识别癌细胞的过程,为癌症的早期诊断提供灵敏和高效的分析方法。
