Following the study of general characteristics of yolk DNA and yolk RNA, a plasmid library of yolk DNA of fertilized eggs was constructed. ss cDNA probes of yolk were obtained by a Dig-labelling procedure and used to screen 200 clones of the plasmid library. 16 clones hybridized with the ss cDNA probes, which indicates that at least part of yolk RNA was transcribed from the yolk DNA. The 16 clones were sequenced and the sequence comparisons with genetic databases revealed that 7 of them was highly similar to those from either Xenopus or other species. These genes encode metabolic enzymes and polyamine transport protein, respectively. So we can speculate that embryotic development may be directed by the yolk DNA. The other 9 clones appeared to be unknown in the genetic databases. They may be the superiovity.genes in yolk.
