To investigate the leukemogenic potential of PML-RARA fusion protein in vivo, the acute promyelocytic leukemia(APL) transgenic mice was generated using transgenic technique. The normal standard of mice blood and bone marrow , the diagnosis standard of transgenic mice in different desease phase was founded. Ac cording to the results in vitro, we concluded that the retinoic acid induced differentiation in APL transgenic mice's bone marrow cell is the same as in APL cell line-NB4 cells, ATRA(100-6 M)can induce differentiation in both of them, it is the basis for us to get more insight into the mechanism of ATRA-induced differentiation in APL cells. Based on this modal, we analysis the gene expression profile in CD34(+) cells from APL transgenic mice in different desease phase. Totally 33 ATRA-modulated genes were identified by cDNA array, these genes are interrelated and interractional ,although the balanced functional network remains unclear , our result provide a very valuble clue for the next step study.
急性早幼粒细胞白血病(APL)具有特异染色体易位t(15;17),导致融合基因PML-RARα纬伞L逋馐笛橐阎っ鱌ML-RARα可使正常细胞发生转化。构建转基因APL小鼠模型不仅可进徊搅私釶ML-RARα在APL中的作用,而且可为筛选新的抗肿瘤药物提供理想的动物模型。
